Abstract:Paper spray ionization has been developed as a direct, fast and low-cost sampling and ionization method for qualitative and quantitative mass spectrometric (MS) analysis of complex mixtures. Analyte ions are generated by applying a high voltage and a small volume (∼10μL) of spray solvent onto a porous substrate. The sample can be preloaded onto the paper or mixed into the spray solution. The geometry of the paper and the method of supplying the necessary internal standard are important factors that affect the ionization efficiency and subsequently the sensitivity and quantitation accuracy of the analytical data. As the cut angle of the paper tip is changed, the spray plume, the total spray current and the electric field intensity at the tip all vary correspondingly, with resulting differences in signal intensity. Sample load is another important factor for obtaining a stable MS signal and accurate quantitative results. The optimal sample load was found to be dependent on the paper size. The dissolution and spray process was also investigated and analyte transfer on paper was shown to be largely associated with bulk solution flow toward the spray tip. The information gathered from these systematic studies provides guidance for the design and optimization of a disposable sample cartridge for paper spray MS, a device which potentially is suitable for fast clinical analysis, especially for point-of-care diagnostics.

Abstract:Paper spray has been developed as an ambient ionization method for direct analysis of biological samples using mass spectrometry. While distinct advantages of paper spray have been demonstrated, especially for quantitative analysis and design of disposable sample cartridges, the need for improvement has also been recognized, especially for the use with miniature mass spectrometers. In this study, we made an improvement to the sampling and ionization by adding a capillary emitter to the paper substrate to produce a paper-capillary spray, which has been shown to have significant, positive impact on the sensitivity and reproducibility for direct mass spectrometry analysis. The paper-capillary devices were fabricated and the effects of the geometry, the treatment of the capillary emitters, as well as the sample disposition methods were characterized. The method’s analytical performance was also characterized for analysis of therapeutic drugs in blood samples. Quantitation of cotinine in blood using a commercial triple quadrupole and sitagliptin (Januvia®) in blood using a desktop Mini 12 ion trap mass spectrometer was also demonstrated.

Abstract:Paper spray is developed as a direct sampling ionization method for mass spectrometric analysis of complex mixtures. Ions of analyte are generated by applying a high voltage to a paper triangle wetted with a small volume (<10 microL) of solution. Samples can be preloaded onto the paper, added with the wetting solution, or transferred from surfaces using the paper as a wipe. It is demonstrated that paper spray is applicable to the analysis of a wide variety of compounds, including small organic compounds, peptides, and proteins. Procedures are developed for analysis of dried biofluid spots and applied to therapeutic drug monitoring with whole blood samples and to illicit drug detection in raw urine samples. Limits of detection of 50 ng/mL (or 20 pg absolute) are achieved for atenolol in bovine blood. The combination of sample collection from surfaces and paper spray ionization also enables fast chemical screening at high sensitivity, for example 100 pg of heroin distributed on a surface and agrochemicals on fruit peels are detectable. Online derivatization with a preloaded reagent is demonstrated for analysis of cholesterol in human serum. The combination of paper spray with miniature mass spectrometers offers a powerful impetus to wide application of mass spectrometry in nonlaboratory environments.

Abstract:We develop a capillary-paper spray (CPS) ion source which allows for sample separation in the capillary and enables rapid and sensitive paper spray (PS) mass spectrometry (MS) analysis of biofluids. The CPS employs a glass capillary to load liquid analytes, vertically standing at the rear of the PS. To further reduce the matrix effect, a nitrocellulose filter membrane is placed between the glass tube and chromatography paper to absorb proteins and other macromolecules, which is beneficial for the detection of the small molecules. Compared with the normal PS method, the CPS method markedly improves spray stability and prolongs analysis duration, and also generates significantly better signal intensities during the analysis of drugs, thus indicating its potential for clinical use. As a proof of concept, quantitative analysis of drugs (metformin hydrochloride and berberine hydrochloride) in serum is performed.

Abstract:Paper spray has been developed as a fast sampling ionization method for direct analysis of raw biological and chemical samples using mass spectrometry (MS). Quantitation of therapeutic drugs in blood samples at high accuracy has also been achieved using paper spray MS without traditional sample preparation or chromatographic separation. The paper spray ionization is a process integrated with a fast extraction of the analyte from the raw sample by a solvent, the transport of the extracted analytes on the paper, and a spray ionization at the tip of the paper substrate with a high voltage applied. In this study, the influence on the analytical performance by the solvent–substrate systems and the selection of the elution methods was investigated. The protein hemoglobin could be observed from fresh blood samples on silanized paper or from dried blood spots on silica-coated paper. The on-paper separation of the chemicals during the paper spray was characterized through the analysis of a mixture of the methyl violet 2B and methylene blue. The mode of applying the spray solvent was found to have a significant impact on the separation. The results in this study led to a better understanding of the analyte elution, on-paper separation, as well as the ionization processes of the paper spray. This study also helps in establishing a guideline for optimizing the analytical performance of paper spray for direct analysis of target analytes using mass spectrometry.

Abstract:Current detection methods for paper-based analytical devices (PADs) rely on spectroscopic and electrochemical properties, which place special requirements on the analyte or need analyte labeling. Here, ion-transmission mass spectrometry (MS) was proposed for coupling with PADs to enable rapid in situ MS analysis of the sample on paper. The sample was analyzed directly on paper via analyte ionization by ions transmitted through the paper, generated by a low-temperature plasma probe. Prior to MS analysis, the sample can be separated by paper electrophoresis or by paper chromatography, among a variety of other features offered by PADs. The versatility of this technique was demonstrated by MS analysis of a paper microarray, a mixture of amino acids, and whole blood doped with drugs on PADs.
Paper Spray Mass Spectrometry-Based Method for Analysis of Droplets in a Gravity-Driven Microfluidic Chip.

Abstract:This work presents a paper spray mass spectrometry-based method, to analyze microdroplets produced in a gravity-driven microchip. Droplets at ambient pressure were passively transferred from the chip to a paper substrate by the capillary wicking effect. Paper spray ionization was then performed for mass spectrometry (MS) analysis of droplet contents. The qualitative and quantitative analytical performances of this technique for single droplets were demonstrated. This manually controlled interface is straightforward, low-cost and simple to implement. Moreover, paper spray ionization MS holds promise in the direct analysis of real biological/chemical microreaction samples because of its tolerance with complex matrices. As a proof-of-concept example, the droplet-based acetylcholine hydrolysis was carried out to demonstrate the validation of our method for the direct analysis of micro-chemical/biological reactions. We also introduced a flow injection analysis (FIA) system combined with our droplet system to generate a concentration gradient. As a result, the microreaction can be performed at different concentrations and kinetic information can be obtained in one sample injection. In conclusion, the combination of a microdroplet chip with paper spray ionization and the introduction of the FIA system and make our droplet-MS scheme a useful platform for monitoring and analyzing organic-phase chemical/biological reactions.

Abstract:Paper spray for direct mass spectrometry analysis is introduced in this chapter. The fundamental study of the mechanism and the characterization of the analytical performance are reviewed for paper spray mass spectrometry (PS-MS). An important aspect in development of paper spray is its capability for quantitation at high precision for analytes in complex samples, which is important for a wide range of applications in many different fields with mandatory criteria in quantitation. Its potential for point-of-care (POC) analysis is discussed with recent implementation with miniature MS analytical systems. Three other ambient ionization methods associated with paper spray, including leaf spray, tissue spray, and extraction spray ionization are also introduced in this chapter.

Abstract:RationalePaper spray (PS) has been developed as a method of choice for point‐of‐care analysis in many real cases, where its applications can be further expanded with delicate high‐throughput design. To achieve this goal, we developed a new PS regime, with the assembly of an induced high voltage into the ion source. Compared with regular DC high voltage, the newly developed setup is capable of high‐throughput, simple configuration and rapid switching between individual papers without complicated electric/mechanic design.MethodsA device of high‐throughput induced PS (IPS) was designed by using a two‐dimensional (2D) rotating platform equipped with a circular glass plate. The paper substrate was placed on the circular glass plate and separated from the electrode. The method avoids physical contact between the electrode and the sample. Charged droplets were generated at the paper tip once an induced high voltage was applied to a wet paper.ResultsA relatively rapid analytical speed of 2.6 s per sample was achieved via IPS‐MS. Rapid quantification of amitriptyline (AMT) in complicated matrices was obtained within 1 min using an isotope internal standard method. Limits of detection for AMt in urine, FBS and blood were calculated to be 1.04, 0.84 and 1.33 ng/mL, respectively. In addition, high‐throughput IPS‐MS can be used for chemical reaction monitoring.ConclusionsWe have demonstrated the versatility of high‐throughput IPS‐MS in ambient ionization, which successfully simplified the experimental installation and facilitated the experimental operation. Therefore, we believe that high‐throughput IPS‐MS analysis will be widely used for discovering drugs and screening reactions, and the present design has the potential for applications in paper chip‐MS analysis.

Abstract:Paper spray ionization mass spectrometry (PSI-MS) is a new technology of mass spectrometry.This method uses low-cost paper as the carrier,and solid or liquid sample is added to a triangular paper substrate.Through the application of high voltage,the target molecules in the high voltage electric field are driven to reach the paper tip,then a charged spray for PSI-MS is generated.This technology combines the characteristics of ambient mass spectrometry (AMS) and electrospray ionization (ESI) technology,and can analyze sample or substance attached to the sample surface in an open atmospheric environment.It has many advantages such as low cost,high throughput,simple,real-time and fast,and it can also simplify the work processes,improving the usability of mass spectrometry equipment.Herein,a general overview of PSI-MS is given in this paper,including the principle,influencing factors (paper substrate,spray solution,elution mode,and internal standard),the improvement and development of the technology,and corresponding applications in various fields (i.e.biological samples,phytochemistry,variety identification,food safety,and environmental analysis) in recent years.Characteristics and advantages of this technology are introduced in details.A number of studies have proved that PSI-MS is a widely used analytical technique with great prospects.Finally,the shortcomings of PSI-MS in terms of accurate quantification,carrier selection and field analysis are discussed,and some ideas for its improvement are put forward.

Abstract:Rationale Accurate measurement of trace compounds in blood samples is important in clinical diagnosis and life science. Ambient ionization mass spectrometry, however, suffers from the matrix effect when dealing with complex samples such as blood. Therefore, it is important to reduce the matrix effects in blood samples. Methods A low-cost and disposable Teflon tube was used as a platform to precipitate the protein in blood. The analytes are extracted into organic solvent, and the precipitated protein can be adsorbed by the chromatography paper inserted. Therefore, the Teflon tube after precipitation can be directly subjected to paper spray ionization mass spectrometry, achieving one-step analysis of blood. Results High sensitivity and satisfactory stability were achieved for pharmaceuticals, acids, and endogenic metabolites in blood. The absolute signal intensities of characteristic product ions of the tested analytes were 8-20 times higher after protein precipitation than those obtained using paper spray. Detection limits and quantitative performance were evaluated for three drugs: carbamazepine, metformin, and tioconazole. In addition, the limits of detection and quantitation were improved 9-14- and 8-12-fold, respectively. Conclusions Protein precipitation coupled to paper spray with a tube and then to mass spectrometry was successfully achieved and applied in the one-step analysis of trace compounds in blood samples. The experimental results showed that this method was sensitive, stable, convenient, and economic for the direct analysis of blood.

Abstract:Paper spray mass spectrometry (PS-MS) is explored as a fast and convenient way for direct analysis of molecules in tissues with minimum sample pretreatment. This technique allows direct detection of different types of compounds such as hormones, lipids, and therapeutic drugs in short total analysis times (less than 1 min) using a small volume of tissue sample (typically 1 mm(3) or less). The tissue sample could be obtained by needle aspiration biopsy, by punch biopsy, or by rubbing a thin tissue section across the paper. There exists potential for the application of paper spray mass spectrometry together with tissue biopsy for clinical diagnostics.

Abstract:The transition of mass spectrometry for clinical analysis is highly desirable, and major progress has been made with direct sampling ionization for operation simplification. High-precision quantitation, however, remains a major challenge in this transition. Herein, a novel method was developed for direct quantitation of biofluid samples, using an extremely simplified procedure for incorporation of internal standards selected against the traditional rules. Slug flow microextraction was used for the development, with conditions predicted by a theoretical model, viz., using internal standards of partition coefficients very different from the analytes and large sample-to-extraction solvent volume ratios. Direct quantitation of drug compounds in urine and blood samples was demonstrated. This development enabled an extremely simplified protocol that is expected to have a significant impact on on-site or clinical analysis.

Abstract:Fast analysis of chemicals in samples of complex mixtures is important for many analytical applications including therapeutic drug monitoring and food safety screening.Based on the paper spray technology developed recently, a high throughput device was developed in this work and used for evaluation of the fast quantitative analysis using paper spray mass spectrometry.The performance of the method was characterized with bovine blood samples spiked with sunitinib and nicotine. At optimized condition, relatively fast analysis speed of 7s/sample was achieved with relatively good LOQs (1 ng/ml) and quantitation.In an application of the method for food screening, LODs better than 1 ng/g were achieved for clenbuterol in beef.

Abstract:Paper spray (PS) ionization is a recently developed ion source that has been used to analyze samples in their native environments at ambient pressure without requiring sample preparation or pre-separation. The design of an ion mobility spectrometer (IMS) coupled with PS ionization can help expand IMS applications to on-site detection of complex liquid samples. We report a paper spray ionization ion mobility spectrometer prototype that consists of a PS source and an ion mobility spectrometer optimized using a numerical simulation. The performance of the design was evaluated by measuring 2, 6-di-tert-butylpyridine (2, 6-DtBP). The mobility spectra of the 2, 6-DtBP exhibited a single-product ion peak with reduced mobility calculated at 1.42 cm(2)∕(V s) and a linear response of 0.1-10 μg∕ml, with an estimated detection limit of 0.05 μg∕ml. The Relative Standard Deviation for 1 μg∕ml was 5.7% over 11 measurements. The highest resolving power (47) was measured for 2, 6-DtBP. Based on these preliminary results, the present PSIMS design is expected to become a tool of choice for the rapid analysis of complex liquid samples.

Abstract:Paper spray MS is part of a cohort of ambient ionization or direct analysis methods that seek to analyze complex samples without prior sample preparation. Extraction and electrospray ionization occur directly from the paper substrate upon which a dried matrix spot is stored. Paper spray MS is capable of detecting drugs directly from dried blood, plasma and urine spots at the low ng/ml to pg/ml levels without sample preparation. No front end separation is performed, so MS/MS or high-resolution MS is required. Here, we discuss paper spray methodology, give a comprehensive literature review of the use of paper spray MS for bioanalysis, discuss technological advancements and variations on this technique and discuss some of its limitations.

Abstract:Paper Spray (PS) is an ambient ionisation technique that is conceptually simple, economical, direct and versatile. In its simplest form, the procedure for operation relies on the application of an electric potential to a triangular-shaped paper substrate with the addition of an extraction/spray solvent to generate charge droplets, without requiring any pneumatic assistance. Despite its promise and rapidly growing popularity, there are some practical challenges associated with PS implementation with scope to enhance its performance further. One such challenge relates to only a small fraction of the Taylor cone expansion being sampled at the MS inlet. In this work we propose a new arrangement for PS, which retains the inherent advantages of this popular technique without introducing additional variables, by using an ambient ion focusing lens that is held at the same potential as the paper substrate.
