Abstract:The title chiral compound, 3-aminocarbonyl-1,2,2-trimethylcyclopentane-1-carboxylic acid, C(10)H(17)NO(3), was prepared from (1R,3S)-camphoric acid. The five-membered ring adopts a conformation which is intermediate between a twist and an envelope. Elongations of the C-C bonds and contractions of the C-C-C bond angles are observed within the five-membered ring. A (1)H NMR spectrum was recorded to assist in distinguishing the amide group from the carboxyl group.

Abstract:The novel reaction of [hydroxyl (((+)-10-camphorsulfonyl)oxy)iodo]benzene (1) with alkenoic acids was reported. When I reacted with various 4-pentenoic acids in CH(3)CN, camphorsulfonylactons were obtained in excellent yields in short times, some had two diastereoisomers, whereas 1 reacted with 5-hexenoic acid, giving middle yield of camphorsulfonylacton; however, 3-butenoic and trans-3-hexenoic acids reacted with l slowly in CH(2)Cl(2), only unsaturated lactones were provided.

Abstract:Borneol and camphor are well known bicyclic monoterpenoids and have been widely used in food, senior aromatic spices and folk medicine in China, India and Japan for more than 2000 years. Although they can be obtained by recrystallization or sublimation, it is still a challenge to prepare high-purity borneol and camphor with high yields. In this study, we introduce a new continuous counter-current chromatography method for the gram-scale preparation of borneol and camphor from Cinnamomum camphora (L.) Presl using heptane/ethyl acetate/methanol/water quaternary solvent system and heptane/methanol/water ternary solvent system. Compared to the heptane/ethyl acetate/methanol/water solvent system, the heptane/methanol/water solvent system was found to possess higher loading capacity, and the heptane/methanol/water (10:7:3, v/v/v) solvent system was found to be the optimal solvent system for the separation of the distillation products of C. camphora. As a result, borneol and camphor were well obtained in a one-step separation with a purity of over 99.9%. In addition, continuous injections method using "on demand" solvent system formulation was used to improve the separation efficiency and reduce solvent consumption, whose results indicated that counter-current chromatography was a powerful and efficient tool to obtain borneol and camphor with high purity.

Abstract:The alkoxylation of camphene with 2-methyl-1,3-propanediol was carried out in the presence of anhydrous macroporous and strong acid cation exchange resins.The effects of various parameters,such as agitation speed, catalyst type,solvent,initial mass ratio of reactants,reaction temperature and reusability of catalysts,were investigated in a 250 mL glass stirred tank reactor in order to optimize the reaction conditions.The conditions for the alkoxylation of camphene with 2-methyl-1,3-propanediol under the catalytic action of anhydrous Lewatit2620 were optimized as follows:agitation speed was 400 r·min-1 ;initial mass ratio of camphene to 2-methyl-1,3-propanediol was 1.5∶1;catalyst loading based on the total mass of reactants was 20%;reaction temperature was 353 K and reaction time was four hours.Under these optimum conditions,the conversion of camphene was 78.80% and the selectivity of isobornyl hydroxyl isobutyl ether was 93.5%.Besides,the catalyst showed good catalytic activity after reusing by eight times.Through lengthening the reaction time,thermodynamic equilibrium constants at 333~370 K were derived.The UNIFAC group contribution method was used to calculate activity coefficients of reactants and to correct the nonideality of reactants.The equilibrium constants decreased with increasing temperature,which demonstrated the alkoxylation of camphene with 2-methyl-1,3-propanediol was exothermic.The equilibriumconstants based on concentration were significantly different from that based on activity,which indicated that it was necessary to take the nonideality of the mixture into account in calculation.Besides,the optimized reaction conditions were also verified in a 5 L double layer glass reaction kettle.From the perspective of conversion of camphene and se-lectivity of isobornyl hydroxyl isobutyl ether,result of the experiment in a 5 L layer glass reaction kettle is almost the same as that of the small-scale experiment,which providing evidence for industrial application by using industrial-grade camphene as the material.

Abstract:In this study, we discussed an efficient approach for the synthesis of camphorquinone from camphor by the continuous reaction of bromination and oxidation. The oxidation of 3-bromocamphor was catalyzed by Fe-porphyrins with air. The catalytic activity of iron-metallated functional porphyrins was investigated under optimization conditions. The results showed that this method was a milder alternative to the reported methods, with a simple, greener procedure in which the advantages were the utilization of air as the oxidant and the use of metalloporphyrins as catalysts.

Abstract:Methoxylation of camphene was carried out in a 500 mL stirred reactor by using cation exchange resins as catalysts. The effects of various parameters including agitation speed, catalyst type, initial reactants amount and reaction temperature on the yield of isobomyl methyl ether were investigated to optimize the reaction conditions. The equilibrium constants based on activities for the methoxylation of camphene catalyzed by Lewatit 2620 from 318.15 to 338.15 K were calculated. The enthalpy changes demonstrated that the reaction was exothermic. Moreover, the kinetics of the methoxylation reaction were also derived and the experimental data were well correlated with a pseudohomogeneous model. Scale-up trial was also performed in a 5 L stirred tank reactor under the optimized conditions, showing good feasibility for industrial production. In addition, the alkoxylation of camphene with other alcohols consisting of 2-6 carbon atoms was studied to explore the impact of the alcohols' structure. (C) 2016 Institution of Chemical Engineers. Abstract:Camphor and carvone exhibit a broad spectrum of biological activity, which determines the prospect of their use as a platform for functionalization to obtain the analogues as potential drugs. The functionalization of camphor and carvone often involves changes to the skeleton of the molecules or their fragmentation. Therefore, in modern medicinal chemistry, research aimed at the development of effective approaches to the synthesis of semisynthetic derivatives of camphor and carvone with preservation of the native framework, demonstrating high biological activity, is in demand. The present work is aimed at the synthesis of new propynyl analogues of camphor and carvone, as well as their conjugates with mono- and disaccharides vi a Cu-catalyzed cycloaddition of acetylenes and azides (Cu- AAC). Alkylation of camphor and carvone with propargyl bromide in the presence of the base KN(SiMe 3 ) 2 –Et 3 B in 1,2-dimethoxyethane (DME) at room temperature provides the target products with yields of 69% and 47%, respectively. Glycosyl azides were obtained by the reaction of peracetylated sugars with trimethylsilyl azide in the presence of SnCl4.The synthesis of 1,2,3-triazolyl glycoconjugates of camphor and carvone with mono- and disaccharides was carried out through Cu(I)-catalyzed 1,3-dipolar cycloaddition of azides to acetylenes (CuAAC) in the presence of Cu and CuSO 4 ·5H 2 O. The structures of the synthesized compounds were determined by NMR. The new propynyl-substituted camphor and carvone, as well as their 1,2,3- triazolylglycoconjugates, can be used as promising building blocks for medicine chemistry.

Abstract:4-Aryl-3,4-dihydrocoumarins are one of the most important structural motifs. Herein, we disclose an enantioselective N -Heterocyclic Carbene catalyzed β-arylation/cyclization of α-bromoenals with 3-aminophenols under mild conditions. The protocol allows for the rapid preparation of 4-aryl-3,4-dihydrocoumarins in acceptable yields with good enantioselectivities. The products of this reaction could be converted into chiral diarylpropanoic acid derivatives without erosion of the enantioselectivity.

Abstract:To avoid the use of peracids oxidant or highly concentrated hydrogen peroxide which is potentially hazardous and explosive, herein, a new route to epsilon-caprolactone was developed in which molecule oxygen was employed as the terminal oxidant. The commercial available N-hydroxyphthalimide and ammonium cerium nitrate were used as the key catalysts for the increased yield of epsilon-caprolactone. For instance, the selectivity of epsilon-caprolactone was obtained 92 % with 85 % conversion of cyclohexanone which was comparable to the strategies using highly concentrated hydrogen peroxide. The sacrificed alcohols were transformed into corresponding ketones which were also valuable chemicals. Furthermore, the efficiency of the alcohols was achieved to unprecedented 52 %. The Baeyer-Villiger oxidation of various other cycloalkanones was also examined. The substituent group effect on the efficiency of sacrificed alcohols was investigated in which weak electron-donating substituent induced nearly quantitative yield of epsilon-caprolactone. The reaction mechanism was studied with the help of electron paramagnetic resonance which indicated the existence of a radical pathway.

Abstract:A practical and chemoselective method is described for the O-methylation of carboxylic acids. Dimethyl malonate, a low toxic and commercially available compound was found to be an effective methylating reagent for a variety of carboxylic acids affording methyl ester products in good to high yields and with excellent chemoselectivity, without the use of strong bases as additives. A mechanism involving the utilization of potassium bromide is tentatively proposed.

Abstract:This article describes the design and optimization of an effective organocatalytic three-component domino alpha-methylenation/Diels-Alder reaction to produce vinyl-substituted cyclohexenecarboxaldehydes in a highly regioselective fashion. In these one-pot transformations, 2-formyl-1,3-butadienes (4) were prepared in situ from alpha,beta-unsaturated aldehydes and formalin and were subsequently trapped with a variety of buta-1,3-dienes. The outcomes of the reactions were dependent on the electronic properties of the dienes. 1-Vinylcyclohexenecarbaldehydes 6 were formed by use of acyclic electron-rich dienes, while the initially formed cycloadducts of 4 with cyclopentadiene underwent Cope rearrangements, leading to the formation of tetrahydro-3H-indene-5-carbaldehyde compounds 7. The mechanisms involved in these reactions were deduced from experimental findings. Furthermore, the method was also extended to one-pot domino methylenation/Diels-Alder reactions of dihydrofurans and dihydropyrans to yield spirocyclic lactols 22. In these reactions, the unstable intermediate hydroxyethyl and hydroxypropyl acroleins behaved as dienophiles, undergoing cycloaddition reactions with dienes with good yields and selectivities. The wide variety of functionalized 1-vinylcyclohex-3-enecarbaldehydes 6, 4-vinylcyclohex-1-enecarbaldehydes 7, and spiro lactols 22 generated through the use of these organocatalytic domino processes as a diversity-oriented synthesis provided useful intermediates for the construction of novel odorants.

Abstract:In this paper, we report a facile synthesis of 2-carbamoyl-2-cyanocyclopropanecarboxylates through a tandem iodosobenzene/tetrabutylammonium iodide-induced oxidative cyclization and a subsequent neighboring group-assisted decarboxylation of the Michael adducts of 2-cyanoacetamides with α,β-unsaturated malonates. This method affords the desired highly functionalized cyclopropanes in moderate to good yields and with excellent diastereoselectivities. In addition, the reaction proceeds smoothly under mild conditions and with good functional group tolerance.

Abstract:Cerium(IV) ammonium nitrate (CAN)-mediated oxidation of methylenecyclobutanes was investigated. Compared with the similar reactions of methylenecyclopropanes, the reaction products were quite different. Instead of the desired ring-enlarged product cyclopentanones, a cyclobutyl ring-intact product, spirocyclobutyl-1,2-dioxethane, was obtained. The structures of these spiroheterocycle-containing compounds are interesting, and the reactions are potentially valuable in both organic synthesis and mechanism research.
Engineering of a borneol dehydrogenase from P. putida for the enzymatic resolution of camphor

Abstract:Several thousand different terpenoid structures are known so far, and many of them are interesting for applications as pharmaceuticals, flavors, fragrances, biofuels, insecticides, or fine chemical intermediates. One prominent example is camphor, which has been utilized since ancient times in medical applications. Especially (-)-camphor is gaining more and more interest for pharmaceutical applications. Hence, a commercial reliable source is needed. The natural sources for (-)-camphor are limited, and the oxidation of precious (-)-borneol would be too costly. Hence, synthesis of (-)-camphor from renewable alpha-pinene would be an inexpensive alternative. As the currently used route for the conversion of alpha-pinene to camphor produces a mixture of both enantiomers, preferably catalytic methods for the separation of this racemate are demanded to yield enantiopure camphor. Enzymatic kinetic resolution is a sustainable way to solve this challenge but requires suitable enzymes. In this study, the first borneol dehydrogenase from Pseudomonas sp. ATCC 17453, capable of catalyzing the stereoselective reduction of camphor, was examined. By using a targeted enzyme engineering approach, enantioselective enzyme variants were created with E-values > 100. The best variant was used for the enzymatic kinetic resolution of camphor racemate, yielding 79% of (-)-camphor with an ee of > 99%. Abstract:Four bifunctional, noncovalent amine-squaramide organocatalysts were prepared from camphor in five steps. The stereochemistry of the prepared catalysts was thoroughly analyzed using various spectroscopic techniques. Their organocatalytic activity was investigated in the Michael addition of acetylacetone to trans-β-nitrostyrene. The addition product was formed in complete conversion and with an enantioselectivity of up to 77% ee. In the reactions catalyzed by the 2-exo-3-endo catalysts, the major (S)-enantiomer was formed, whereas in the presence of 2-endo-3-endo catalysts, the (R)-enantiomer was formed as the major product.

Abstract:2-Methylenecyclobutanones (2-MCBones), used to be difficult to access, but can now be easily achieved by a green and stereospecific Ca(OH)(2)-catalyzed direct and simple aldol condensation of cyclobutanone and aldehydes under mild conditions. The obtained (E)-2-MCBones should be a class of potentially useful building blocks in synthesis as they could readily undergo an interesting (PhSe)(2)-catalyzed Baeyer-Villiger (BV) oxidation with H2O2 at room temperature to give the versatile 4-methylenebutanolides. Mechanistic studies revealed that the BV reaction most possibly proceeded via the initial formation of benzeneseleninoperoxoic anhydride [PhSe(O)O](2)O, which then converted to benzeneseleninoperoxoic acid PhSe(O)OOH as the active oxidant, followed by its selective addition to the C=O bond of 2-MCBones and then a selective C-C bond cleavage and rearrangement to give 4-methylenebutanolides.
