Abstract:Bacteria infection have been a major threat to human health. Especially, Gram-negative (G-) bacteria infection have been an increasing problem worldwide. The overuse of antibiotics leads to an emergence of drug resistance, and then the development of novel antimicrobial agents is important, particularly against G- bacteria. Nanozymes use reactive oxygen species (ROS) to kill bacteria, reducing the risk of bacterial resistance and providing new opportunities to meet the challenges of strain selectivity. Here, we synthesized NiCo layered double hydroxides (LDHs) nanozyme, which exhibit selective antibacterial activity based on their peroxide-like (POD-like) activity. To obtain the highest antibacterial activity, the POD-like activity of NiCo LDHs nanozyme was further optimized though tuning the ratio of nickel and cobalt, and Ni4Co6 LDHs showed the highest POD activity and antibacterial activity. More importantly, Ni4Co6 LDHs can achieve selective sterilization of G- bacteria attributing to their electrostatic adsorption and hydrophilic interactions with the bacterial cell wall. Animal experiments further indicated that the healing of G- bacteria -infected wounds was effectively promoted without damaging their normal biological tissues. In conclusion, we provide a selective antibacterial agent through a simple strategy, which provides a new direction for the application of nanozymes.

Abstract:Bacterial infection, tissue hypoxia, and inflammatory and oxidative stress are several key problems in wound healing of chronic infections. Herein, a multi-enzyme-like activity exhibiting multifunctional hydrogel made up of mussel-inspired carbon dot reduced-Ag (CDs/AgNPs) and Cu/Fe-nitrogen-doped carbon (Cu,Fe-NC) was designed. Due to the loss of glutathione (GSH) and oxidase (OXD)-like activity of the nanozyme (decomposes O2 to generate a superoxide anion radical (O2˙-) and hydroxyl radical production (˙OH)), the multifunctional hydrogel exhibited excellent antibacterial performance. More importantly, during the bacterial elimination within the inflammatory phase of wound healing, the hydrogel could act as a catalase (CAT)-like agent to supply adequate O2 by catalyzing intracellular H2O2 for hypoxia abatement. The catechol groups on the CDs/AgNPs endowed them with the dynamic redox equilibrium properties of phenol-quinones, thus providing the hydrogel with mussel-like adhesion properties. The multifunctional hydrogel was shown to excellently promote bacterial infection wound healing and maximize the efficiency of nanozymes.

Bioinspired construction of ATP/Co-Al-Zn LDH nanozyme with enhanced peroxidase-mimic performance for efficient bactericidal activity through membrane disruption

Abstract:In recent years, overuse of antibiotics has led to emerging antibiotic-resistant strains of bacteria. Consequently, creating new, highly productive antibacterial agents is crucial. In this work, we synthesized copper-aluminum-zinc layered double hydroxide (Co-Al-Zn LDH) and modified it using adenosine triphosphate. After characterization, the enzyme-like activity of the prepared particles was evaluated. The results indicated peroxidase-mimic performance of ATP/Co-Al-Zn LDH with Km values of 0.38 mM and 1.69 mM for TMB (3,3',5,5'-tetramethylbenzidine) and hydrogen peroxide (H2O2), respectively, which were lower than that of horseradish peroxidase. The highest peroxidase-like activity of ATP/Co-Al-Zn LDH was achieved at 20 °C, pH 4, with a 1.02 mg/mL catalyst, 231 μM TMB, and 1.9 mM H2O2. The bactericidal activity of the developed nanozyme was studied against E. coli and S. aureus. The peroxidase-mimic nanozyme decomposes H2O2 and generates free radicals to kill bacteria in vitro. The minimum inhibitory concentration (MIC) of ATP/Co-Al-Zn LDH was 15 μg/mL and 20 μg/mL for S. aureus and E. coli, respectively. The morphological characteristics of the nanozyme-treated bacterial cells showed dramatic changes in bacterial morphology. Our results revealed higher antibacterial activity of ATP/Co-Al-Zn LDH against S. aureus. Therefore, the developed nanozyme could serve as a substitute for conventional antibiotics.

Abstract:As common reactive oxygen species, H2O2 is widely used for bacterial inactivation and wound disinfection. However, the concentrations used are always higher than physiological levels, which frequently result in potential toxicity to healthy tissue and even delay wound healing. Here we report highly efficient nanozyme hybrids that are capable of activating biologically relevant concentrations of H2O2 for defending bacterial infections. The integration of AuNPs with ultrathin graphitic carbon nitride (g-C3N4) provides excellent peroxidase-activity, which can catalyze the decomposition of H2O2 to OH radicals much more efficiently, allowing the use of bio-safety levels of H2O2 for the first time. Furthermore, our system not only exhibits striking bactericidal performance against both DR Gram-negative and DR Gram-positive bacteria, but also shows high efficiency in breaking down the existing DR-biofilms and prevented formation of new biofilms in vitro. More importantly, in vivo experiments indicate that our system could significantly prevent bacterial infections and accelerate the healing rate of wounds.

Abstract:Bacterial infection is one of the main causes of delayed wound healing. Hence, the development of antimicrobial dressings with good biocompatibility has high clinical significance and social value. The natural macromolecule, serum albumin, has superior biocompatibility, low immunogenicity, and a good drug-carrying capacity. In this study, human serum albumin (HSA) was modified with methacrylic anhydride to obtain photo-crosslinking properties. Then, the photo-crosslinking HSA was applied to produce homogenized microspheres (HMS) through the nitrogen elution-assisted microfluidic technique and ultraviolet light. Effective loading of vascular endothelial growth factor (VEGF) was achieved by utilizing the adsorption of HSA onto zinc ions and their crosslinking with His-tagged VEGF. The prepared HMS-Zn@VEGF exhibited broad-spectrum antibacterial activity and simultaneously exerted proangiogenic effects in vitro , effectively promoting the healing of infected wounds in rats. In vivo experiments using rats showed that HMS-Zn@VEGF promoted the deposition of collagen around the wound, improved the local inflammatory response, and promoted angiogenesis. In conclusion, our prepared HMS-Zn@VEGF microspheres have good biocompatibility, antimicrobial properties, and angiogenic ability, which can be used for infected wound healing.

Abstract:Colloidal surface engineering is of particular importance to impart modular functionalities to the colloidal systems. Here, a layer of Mn/Ni layered hydroxides (Mn/Ni(OH)x LDHs) can be successfully coated on various colloidal particles, such as silica spheres, silica rods, ferrite nanocrystal supraparticles, as well as FeOOH nanorods. Such layered hydroxides have intrinsic oxidase‐mimetic activities, as demonstrated by catalytic oxidation of tetramethyl benzidine in the presence of oxygen. Furthermore, Mn/Ni(OH)x LDHs structure seems to capture bacteria (both Gram positive and Gram negative) and exhibit antibacterial properties in vitro. Moreover, local delivery of Mn/Ni‐LDH structure fights against infection and reverses delayed wound healing procedures in mice models. Importantly, such hierarchical structures may have strong adhesive properties to the bacteria, which may maximize the contact between Mn/Ni(OH)x LDHs and the bacteria's surface. Overall, the present versatile colloidal surface engineering strategy will bring new insights in the field of antibiotics for its high efficiency toward antibacterial activity.

Abstract:Abstract Metal-based nanomaterials usually have broad-spectrum antibacterial properties, low biological toxicity and no drug resistance due to their intrinsic enzyme-like catalytic properties and external field (magnetic, thermal, acoustic, optical and electrical) responsiveness. Herein, iron oxide (Fe3O4) nanoparticles (IONPs) synthesized by us have good biosafety, excellent photothermal conversion ability and peroxidase-like catalytic activity, which can be used to construct a photothermal-enzymes combined antibacterial treatment platform. IONPs with peroxide-like catalytic activity can induce H2O2 to catalyze the production of •OH in a slightly acidic environment, thus achieving certain bactericidal effects and increasing the sensitivity of bacteria to heat. When stimulated by near-infrared light, the photothermal effect could destroy bacterial cell membranes, resulting in cleavage and inactivation of bacterial protein, DNA or RNA. Meanwhile, it can also improve the catalytic activity of peroxidase-like and promote IONPs to catalyze the production of more •OH for killing bacteria. After IONPs synergistic treatment, the antibacterial rate of Escherichia coli and Staphylococcus aureus reached nearly 100%. It also has an obvious killing effect on bacteria in infected wounds of mice and can effectively promote the healing of S. aureus-infected wounds, which has great application potential in clinical anti-infection treatment.

Abstract:Developing injectable hydrogel dressings with multifunctional properties, including antibacterial, antioxidant ability, and good mechanical property to treat infected full-thickness skin wound is of particular importance in clinical application. Incorporation of metal ions, such as Zn2+, Ca2+, and Fe2+ confer the hydrogels with unique properties, including conductivity, bioactivity, and tunability. In this study, a multifunctional near infrared (NIR) active hydrogel, named Alg-HA-Zn-PDA, was fabricated via the incorporation of polydopamine (PDA) nanoparticles into hyaluronic acid (HA) and alginate (Alg) crosslinked by Zn2+. This hydrogel exhibited high porosity, good mechanical property, and high water content which is essential requirement in the adsorption of exudates during the healing process. PDA endowed the hydrogels with antioxidant activity, hemostatic ability, and made the hydrogels NIR active. Zn2+ could contribute in antibacterial and healing effects of the hydrogels. The antibacterial activity of Alg-HA-Zn-PDA, with and without NIR irradiation against Escherichia coli (E.coli) and Staphylococcus aureus (S. aureus), showed that the NIR light irradiation could result in heat generation and destroyed the bacteria. Furthermore, in vivo treatment in a S. aureus-infected full-thickness skin defect model demonstrated that the Alg-HA-Zn-PDA hydrogel possess excellent wound closure capacity of the hydrogels in the presence of NIR irradiation. In summary, the antibacterial, hemostatic, and antioxidant hydrogels is an excellent candidate as wound dressing for the repairing of full-thickness skin defects.

Customized A-D-A type molecule to construct a nitric oxide nanogenerator with enhanced antibacterial activity for infected wound healing

Abstract:Bacterial infections pose an increasingly serious threat to global health due to the development of drug-resistant strains. Developing a method to efficiently kill bacteria and promote tissue repair is imperative to decrease the damage from bacterial infection, especially infected wounds. Herein, a biofriendly and light-controlled nitric oxide (NO) generator HFB with simultaneous bacterial killing and wound repair properties is reported based on a tailored light-responsive molecule F(EIBC)2. HFB demonstrates an appropriate photothermal conversion efficiency of 33.4% and type I reactive oxygen species (˙OH and H2O2) generation capability to simultaneously trigger NO generation and potently kill bacteria. Furthermore, HFB can effectively eradicate mature bacterial biofilms with the aid of favorable permeability of NO. Additionally, HFB effectively eradicates Staphylococcus aureus in infected wounds of living mice and accelerates healing via NO-induced angiogenesis and collagen deposition. Owing to the encapsulated human serum albumin (HSA), heavy metal-free feature, and synergistic killing mechanism, HFB exhibits good biosafety to surrounding tissue and major organs. This work provides a novel dual-functional photo-responsive molecule and a potential light-controlled release platform for the treatment of bacterial infections.

Abstract:Bacterial infection can hinder the infected wound healing process. Because of the growing drug-resistance bacteria, there is an urgent desire to develop alternative antibacterial strategies to the antibiotics. Herein, the quaternized chitosan coated CuS (CuS-QCS) nanozyme with peroxidase (POD)-like activity was developed through a facile biomineralized approach for synergistic efficient antibacterial therapy and wound healing. The CuS-QCS killed bacteria by the electrostatic bonding of positive charged QCS with bacteria and releasing Cu 2+ to damage bacterial membrane. And importantly, CuS-QCS nanozyme exhibited higher intrinsic POD-like activity, which converted H 2 O 2 with low concentration into highly toxic hydroxyl radical (OH) for the elimination of bacteria by oxidative stress. Through cooperation of POD-like activity, Cu 2+ and QCS, CuS-QCS nanozyme exhibited excellent antibacterial efficacy of approximate 99.9 % against E. coli and S. aureus in vitro . In addition, the QCS-CuS was successfully used to promote the healing of S. aureus infected wound with good biocompatibility. This synergistic nanoplatform presented here shows great potential applications in the field of wound infection management.

Iron Single-Atom Nanozyme with Inflammation-Suppressing for Inhibiting Multidrug-Resistant Bacterial Infection and Facilitating Wound Healing

Abstract:Infection with drug-resistant bacteria and the formation of biofilms are the main factors contributing to wound healing insufficiency. Antibacterial agents with enzyme-like properties have exhibited considerable potential for efficient eradication of drug-resistant microorganisms due to their superior sensitivities and minimal side effects. In this work, we prepared a kind of Fe-centered single-atom nanozyme (Fe-SAzyme) with high biocompatibility and stability via a facile one-pot hydrothermal method, which was suitable for the treatment of wounds infected with drug-resistant bacteria. The Fe-SAzyme exhibited remarkable peroxidase-like catalytic activities, catalyzing the conversion of hydrogen peroxide (H2O2) to highly toxic hydroxyl radicals (•OH), which could not only damage bacterial cells but also inhibit, disrupt, and eradicate the formation of bacterial biofilms. Thus, Fe-SAzyme demonstrated a broad-spectrum antibacterial performance capable of effectively eliminating multidrug-resistant bacteria. The coexistence of ferrous (Fe2+) and ferric (Fe3+) ions in Fe-SAzyme conferred the nanozyme with anti-inflammatory activity, effectively suppressing excessive inflammation. Meanwhile, Fe-SAzyme could significantly downregulate inflammatory cytokines tumor necrosis factor-α and interleukin-1β and upregulate growth factors VEGF and epidermal growth factor, which can prevent bacterial infection, mitigate inflammation, promote fibroblast proliferation, and improve wound closure. Thus, Fe-SAzyme had shown favorable therapeutic efficiency in promoting bacteria-infected wound healing. This study provides Fe-SAzyme as a promising candidate for the development of new strategies to treat multidrug-resistant bacterial infections.
pH-responsive double-enzyme active metal–organic framework for promoting the healing of infected wounds

Abstract:The abuse of antibiotics accelerates the spread and evolution of drug-resistant bacteria, which seriously threatens human health. Hydroxyl radicals (•OH) are generated by peroxidase in the presence of H2O2, which is strongly oxidizing and can effectively kill bacteria. However, high production costs and poor stability limit the clinical use of natural enzymes. "Nanozyme" is a general term for nanomaterials with catalytic activity similar to that of biological enzymes. Compared to biological enzymes, nanozymes have the advantages of low cost, facile preparation, and easy storage, making them a good choice for the development of antibacterial agents. Here, a nickel-based metal-organic framework (Ni-MOF) with dual enzymatic activity that switches depending on the pH environment was studied. In a slightly acidic environment, Ni-MOF can react with hydrogen peroxide to produce hydroxyl radicals that kill bacteria; in a neutral environment, Ni-MOF instead removes excessive reactive oxygen species (ROS) and promotes the transformation of macrophages into M2 macrophages. Compared to most nanozymes, Ni-MOF has unique electrical conductivity and better biosafety. The results of animal experiments show that Ni-MOF can not only treat infected wounds but also promote the healing of acute wounds and exhibits great clinical application potential.

Self-Driven Electron Transfer Biomimetic Enzymatic Catalysis of Bismuth-Doped PCN-222 MOF for Rapid Therapy of Bacteria-Infected Wounds.

Abstract:In this work, a biomimetic nanozyme catalyst with rapid and efficient self-bacteria-killing and wound-healing performances was synthesized. Through an in situ reduction reaction, a PCN-222 metal organic framework (MOF) was doped with bismuth nanoparticles (Bi NPs) to form Bi-PCN-222, an interfacial Schottky heterojunction biomimetic nanozyme catalyst, which can kill 99.9% of Staphylococcus aureus (S. aureus). The underlying mechanism was that Bi NP doping can endow Bi-PCN-222 MOF with self-driven charge transfer through the Schottky interface and the capability of oxidase-like and peroxidase-like activity, because a large number of free electrons can be captured by surrounding oxygen species to produce radical oxygen species (ROS). Furthermore, once bacteria contact Bi-PCN-222 in a physiological environment, its appropriate redox potential can trigger electron transfer through the electron transport pathway in bacterial membranes and then the interior of the bacteria, which disturbs the bacterial respiration process and subsequent metabolism. Additionally, Bi-PCN-222 can also accelerate tissue regeneration by upregulating fibroblast proliferation and angiogenesis genes (bFGF, VEGF, and HIF-1α), thereby promoting wound healing. This biomimetic enzyme-catalyzed strategy will bring enlightenment to the design of self-bacterial agents for efficient disinfection and tissue reconstruction simultaneously.
Polymetallic Hybrid Nanoplatform with Hyperthermia‐Amplified Dual Enzyme‐Like Activities for Efficient Speeded‐Up Bacterially Infected Wound Healing

Abstract:Spike‐like [Cu(OH)2@Au@Co(OH)2] nanoparticles (CAC NPs) display near‐infrared (NIR) irradiation‐amplified oxidase and peroxidase biocatalytic activities that increase ROS generation and promote bactericidal effects in vitro and in vivo. Antibiotic‐independent artificial nanozymes exhibit excellent biocatalytic activity, and can generate sufficient reactive oxygen species (ROS) for potential applications in the field of anti‐infective therapy. However, single nanozymes often catalyze the production of ROS with low yield and low utilization, resulting in low antibacterial activity. To address the above refractory concern, a spike‐like surface nanoparticles (CAC NPs) platform is built that possesses intrinsic dual enzyme‐like (oxidase and peroxidase‐like) activities and photothermal conversion capacity with near‐infrared irradiation (NIR‐I) to quickly capture and effectively eliminate the bacteria and biofilm. Upon NIR‐I irradiation, CAC NPs have exerted an excellent biocatalytic antibacterial effect through a hyperthermia‐amplified dual enzyme‐like activities strategy. As predicted, the results show CAC NPs can eradicate bacteria in the subcutaneous tissues of mice and promote the healing of inflamed tissues with no significant side effects in vivo. Collectively, this work reveals the potential of NIR‐assisted dual enzyme‐like activities for antimicrobial therapy and provides broad prospects for antimicrobial drugs and strategies.

Abstract:Bacterial wound infections are a serious threat due to the emergence of antibiotic resistance. Herein, we report an innovative hybrid nanozyme independent of antibiotics for antimicrobial wound healing. The hybrid nanozymes are fabricated from ultra-small Au NPs via in-situ growth on metal-organic framework (MOF)-stabilised Fe 3 O 4 NPs (Fe 3 O 4 @MOF@Au NPs, FMA NPs). The fabricated hybrid nanozymes displayed synergistic peroxidase (POD)-like activities. It showed a remarkable level of hydroxyl radicals (·OH) in the presence of a low dose of H 2 O 2 (0.97 mM). Further, the hybrid FMA nanozymes exhibited excellent biocompatibility and favourable antibacterial effects against both Gram-negative (Escherichia coli) and Gram-positive ( Staphylococcus aureus ) bacteria. The animal experiments indicated that the hybrid nanozymes promoted wound repair with adequate biosafety. Thus, the well-designed hybrid nanozymes represent a potential strategy for healing bacterial wound infections, without any toxic side effects, suggesting possible applications in antimicrobial therapy.

H2O2/Acid Self-Supplying Double-Layer Electrospun Nanofibers based on ZnO2 and Fe3O4 nanoparticles for Efficient Catalytic Therapy of Wound Infection

Abstract:Catalytic therapy based on nanozymes is a promising method for the treatment of bacterial infections. Nanozymes with peroxidase (POD)-like activity can convert hydrogen peroxide (H2O2) to produce reactive oxygen species (ROS) for killing bacteria, especially those resistant to antibiotics. However, the therapeutic efficacy of catalytic therapy is usually restricted by a limited amount of H2O2 and a weak acidic environment in infected tissues. To solve these issues, we prepared polyvinyl alcohol (PVA)-polyacrylic acid (PAA)-iron oxide (Fe3O4)/polyvinyl alcohol (PVA)-zinc peroxide (ZnO2) double-layer electrospun nanofibers (PPF/PZ NFs) to provide both H2O2 and acid. In this design, PVA serves as the carrier for ZnO2 nanoparticles (NPs), Fe3O4 NPs, and PAA. The double-layer structure of nanofibers can spatially separate the PAA and ZnO2 to avoid their reaction with each other during preparation and storage. While in the wet wound bed, PVA can dissolve and PAA can provide H+ to promote the generation of H2O2 and subsequent conversion to ▪OH for bacteria killing. In vitro experimental results demonstrated that PPF/PZ NFs can reduce the methicillin-resistant Staphylococcus aureus (MRSA) by 3.1 log (99.92%). Moreover, PPF/PZ NFs can efficiently treat MRSA infection in mice wound model and promote wound healing with negligible toxicity to animals, indicating their potential use as "plug-and-play" antibacterial wound dressings. This work provides a novel strategy for the construction of double-layer electrospun nanofibers as catalytic wound dressings with H2O2/acid self-supplying properties for the efficient treatment of bacterial infections.

Abstract:As a broad-spectrum antimicrobial material, ZnO nanoparticles (NPs) offer a novel approach to infected wounds in the clinic; however, it is very necessary to improve the antimicrobial performance of ZnO to satisfy the clinic requirements. In this work, a new antimicrobial hybrid ZnO@PDA/GL13K was prepared by attaching the antimicrobial GL13K peptide on the surface of ZnO NPs via polydopamine (PDA) as a covalent bonding agent, which can enhance the reactive oxygen species (ROS) production and damage the cell wall, showing superior antimicrobial efficacy. Besides, the ZnO@PDA/GL13K hybrid can promote the healing of bacterial infected wounds, with the wound area only remaining 1.8% on day 9, about 2 times smaller than that of the ZnO group. Moreover, ZnO@PDA/GL13K also showed good biocompatibility, and no side effect on normal tissue and organs was found, implying that the antimicrobial hybrid may possess prospective application in biomedical fields.
Engineering Platelet Membrane‐Coated Bimetallic MOFs as Biodegradable Nanozymes for Efficient Antibacterial Therapy

Abstract:Abstract Nanocatalytic‐based wound therapeutics present a promising strategy for generating reactive oxygen species (ROS) to antipathogen to promote wound healing. However, the full clinical potential of these nanocatalysts is limited by their low reactivity, limited targeting ability, and poor biodegradability in the wound microenvironment. Herein, a bio‐organic nanozyme is developed by encapsulating a FeZn‐based bimetallic organic framework (MOF) (MIL‐88B‐Fe/Zn) in platelet membranes (PM@MIL‐88B‐Fe/Zn) for antimicrobial activity during wound healing. The introduction of Zn in MIL‐88B‐Fe/Zn modulates the electronic structure of Fe thus accelerating the catalytic kinetics of its peroxidase‐like activity to catalytically generate powerful ROS. The platelet membrane coating of MOF innovatively enhanced the interaction between nanoparticles and the biological environment, further developing bacterial‐targeted therapy with excellent antibacterial activity against both gram‐positive and gram‐negative bacteria. Furthermore, this nanozyme markedly suppressed the levels of inflammatory cytokines and promoted angiogenesis in vivo to effectively treat skin surface wounds and accelerate wound healing. PM@MIL‐88B‐Fe/Zn exhibited superior biodegradability, favourable metabolism and non‐toxic accumulation, eliminating concerns regarding side effects from long‐term exposure. The high catalytic reactivity, excellent targeting features, and biodegradability of these nanoenzymes developed in this study provide useful insights into the design and synthesis of nanocatalysts/nanozymes for practical biomedical applications.

A synergistically antimicrobial and antioxidant hyaluronic acid hydrogel for infected wounds

Abstract:Bacterial infections during wound healing impede the healing process and trigger local or systemic inflammatory reactions. Consequently, there is an urgent need to develop a new material with antimicrobial and antioxidant properties to promote infected wound healing. A synergistically antimicrobial and antioxidant hyaluronic acid hydrogel (HMn) is prepared by employing MnO2 nanosheets into 4ARM-PEG5000-SH crosslinked methacrylated hyaluronic acid (HAMA) network. The coordination between sulfhydryl groups of 4ARM-PEG5000-SH and MnO2 nanosheets ensures entrapment of the nanosheets within the hydrogel, while the interaction between 4ARM-PEG5000-SH and HAMA results in facile gelation through thiol-ene click reaction. MnO2 nanosheets exhibit strong photothermal properties and reactive oxygen species (ROS) scavenging abilities, while hyaluronic acid promotes wound healing. When subjected to near-infrared (NIR) irradiation, the HMn achieves a bactericidal rate of 95.24 % for Staphylococcus aureus and nearly 100 % for Escherichia coli. In animal experiments, treatment with the HMn under NIR irradiation results in the best wound healing outcomes. Both in vitro and vivo biocompatible assays demonstrate that the HMn has rarely cell cytotoxicity and tissue damage. The HMn is easy to prepare and has good biocompatibility as well as efficient antibacterial and antioxidant properties, providing a novel method for the treatment of infected wounds.
An injectable chitosan-based hydrogel incorporating carbon dots with dual enzyme-mimic activities for synergistically treatment of bacteria infected wounds

Abstract:Bacterial infections pose a serious threat to human health, and the emergence of superbugs and the growing antibiotic resistance phenomenon have made the development of novel antimicrobial products. In this paper, an ultrasmall Cu, N co-doped carbon dots (CDs-Cu-N) with excellent peroxidase mimic activity and enhanced catalase mimic activity was successfully prepared and anchored to an injectable chitosan (CS)-based hybrid hydrogel. As expected, the CDs-Cu-N-H2O2-CS hybrid hydrogel maintains the excellent enzyme-mimicking properties of CDs-Cu-N and shows superior antibacterial property, which has been proven to effectively promote the healing of S. aureus-infected wounds with good biocompatibility. Benefitting from the dual-enzyme-mimic activity of CDs-Cu-N, the hybrid hydrogel not only can catalyze the generation of highly toxic ROS from low concentration of H2O2 to inhibit the bacterial infections, but also can significantly promote the wound tissue repair and regeneration by improving the anoxic microenvironment and promoting neovascularization. In addition, this hybrid hydrogel also possessed excellent injectability and moldability. It can adapt to various the irregular shapes of acute wounds, maintaining a moist and safe microenvironment while prolonging the action time of nanozyme on wounds, thus promoting wound healing. This injectable hybrid hydrogel shows great potential applications in the field of wound infection management.
