Comparative response to PDT with methyl-aminolevulinate and temoporfin in cutaneous and oral squamous cell carcinoma cells

Abstract:Cutaneous and Head and Neck squamous cell carcinoma (CSCC, HNSCC) are among the most prevalent cancers. Both types of cancer can be treated with photodynamic therapy (PDT) by using the photosensitizer Temoporfin in HNSCC and the prodrug methyl-aminolevulinate (MAL) in CSCC. However, PDT is not always effective. Therefore, it is mandatory to correctly approach the therapy according to the characteristics of the tumour cells. For this reason, we have used cell lines of CSCC (A431 and SCC13) and HNSCC (HN5 and SCC9). The results obtained indicated that the better response to MAL-PDT was related to its localization in the plasma membrane (A431 and HN5 cells). However, with Temoporfin all cell lines showed lysosome localization, even the most sensitive ones (HN5). The expression of mesenchymal markers and migratory capacity was greater in HNSCC lines compared to CSCC, but no correlation with PDT response was observed. The translocation to the nucleus of β-catenin and GSK3β and the activation of NF-κβ is related to the poor response to PDT in the HNSCC lines. Therefore, we propose that intracellular localization of GSK3β could be a good marker of response to PDT in HNSCC. Although the molecular mechanism of response to PDT needs further elucidation, this work shows that the most MAL-resistant line of CSCC is more sensitive to Temoporfin.

The problem that this paper attempts to solve is to evaluate the differences in the photodynamic therapy (PDT) responses of two photosensitizers (methyl - aminolevulinate, MAL and temoporfin) in cutaneous squamous cell carcinoma (CSCC) and head and neck squamous cell carcinoma (HNSCC) cells, and to explore the relationships between these differences and the epithelial - mesenchymal transition (EMT) process and the activation of the GSK3β/NF - κβ signaling pathway. Specifically, the main objectives of the study include: 1. **Compare the responses of different cell lines to MAL - PDT and Temoporfin - PDT**: - Select CSCC cell lines (A431 and SCC13) and HNSCC cell lines (HN5 and SCC9), and evaluate their sensitivities to the two photosensitizers, MAL and Temoporfin, through MTT assays. 2. **Analyze cell proliferation, migration ability, and EMT marker expression**: - Evaluate the proliferation and migration abilities of different cell lines by detecting the cell proliferation index, colony - forming ability, and wound - healing assay. - Analyze the expression of EMT markers such as E - cadherin, β - catenin, N - cadherin, vimentin, and snail using immunofluorescence and Western Blot techniques. 3. **Study the expression and localization of the GSK3β/NF - κβ signaling pathway**: - Analyze the subcellular localization and expression levels of GSK3β and NF - κβ in different cell lines, and explore their relationships with the PDT response. 4. **Explore the subcellular localization of photosensitizers and ROS generation**: - Use fluorescence microscopy to observe the subcellular localizations of PpIX (generated by MAL) and Temoporfin, and understand their distributions in different cell lines. - Measure the ROS generation level to evaluate the intracellular oxidative stress response under different PDT treatment conditions. Through the above - mentioned research, the authors hope to gain an in - depth understanding of the response mechanisms of different types of squamous cell carcinomas to PDT, thereby providing a more rational basis for the selection of photosensitizers in clinical treatment.
