Abstract:The changes in rice starch fine structures and functional properties induced by drought, elevated CO2 (eCO2) and combined treatments remain to be elucidated comprehensively. In this study, starch molecular structures were characterized by size-exclusion chromatography and fluorophore-assisted carbohydrate electrophoresis, and the data from amylopectin chain-length distributions was fitted with a mathematical model to obtain parameters for statistical analysis. Starch crystalline structures, thermal and pasting properties of rice flour were also analyzed. All the treatments decreased the seed setting rate and increased chalky grains, reflecting the abnormal starch accumulation. However, eCO2 partially increased the seed setting rate and chalky grains under drought stress. The stress-treated starch samples had shorter amylopectin short and intermediate chains, the lower degree of ordered structures, but similar amylose content and relative crystallinity. This led to lower onset gelatinization temperature and paste viscosities, which may be responsible for the worse sensory quality of rice grains.

Abstract:The evolution of the starch fine structure during growth and its impact on the gelatinization behavior of cassava starch (CS) was investigated by isolating starch from South China 6068 (SC6068) cassava harvested from the 4th to 9th growth period. During growth, the short-range ordered structure, crystallinity as well as particle size distribution of starch were increased. Meanwhile, the starch molecular size and amylopectin (AP) proportion increased, while the proportion of amylose (AM) exhibited a decreasing tendency. The chains of short-AM (X - 100-1000) were mainly significantly reduced, whereas the short and medium-AP chains (X - 6-24) had the most increment in AP. The solubility, thermal stability, shear resistance, and retrogradation resistance of starch were enhanced after gelatinized under the influence of the results mentioned above. This study presented a deeper insight into the variation of starch fine structure during growth and its influence on gelatinization behavior, which would provide a theoretical basis for starch industrial applications.
In Vitro Fecal Fermentation Characteristics of Mutant Rice Starch Depend More on Amylose Content Than Crystalline Structure

Abstract:To obtain the relation between rice starch features and fermentation characteristics, rice starches with various polymorphic types and apparent amylose contents were subjected to in vitro fecal fermentation. Gas and short-chain fatty acid production was evaluated as a function of fermentation time, and the microbial responses were monitored by 16S rRNA sequencing technique at the end of fermentation. Regardless of polymorphic type, three high-amylose mutant rice starches (i.e., GM03, A-type; BP577, B-type; Wx21TT, C-type) displayed significantly slower fermentation rate during the first 12 h and higher final butyrate yield (17.6-17.9 mM) compared to the A-type normal starches (9311 and Wx22TT), and promoted the proliferation of Roseburia. However, A-type normal rice starches presented higher propionate production, and increased the growth of Bacteroides and Megamonas. The principal component and redundancy analyses indicated that three high-amylose mutant rice starches showed similar abundance and migration of microbial communities, and the apparent amylose content was closely correlated with the abundance of their five key amplicon sequence variants. Our results demonstrated that amylose content might be a controlling factor in determining the fermentation properties of rice starches than crystalline structure.
Physicochemical properties and digestibility of endosperm starches in four indica rice mutants.

Abstract:Structural, physicochemical characteristics and digestibility of endosperm starches from four mutants (GM01, 03-05) and their parent Guangluai 4 (GLA4) were characterized. GM05 had a little higher apparent amylose content (AAC) and showed little difference in starch and digestion properties from GLA4. However, GM03 and GM04 with increased amount of chalkiness and partial translucent region in the endosperm displayed a distinct starch and digestion properties, which had the RS of 7.9 and 8.4%, respectively. GM03 and GM04 had higher AAC (33-35%), lower degree of crystallinity, lower gelatinization temperature and pasting viscosities, and more amount of B1 (DP 12-24) and B2 (DP 25-36) chains and less amount of B3 chains (DP ≥ 37) in amylopectin. AAC and the amount of B1 chains had positive correlation with RS, but the amount of B3 chains had negative correlation with RS. The results of this study may be applied to design RS by selecting rice germplasm with high AAC and high amount of B1 chains (DP 12-24) of amylopectin.
Relationships among Genetic, Structural, and Functional Properties of Rice Starch

Abstract:We determined the relationships among the structural properties, in vitro digestibility, and genetic factors in starches of 14 rice cultivars. Weight-based chain-length distributions in amylopectin ranged from 18.07% to 24.71% (fa, DP 6-12), 45.01% to 55.67% (fb1, DP 13-24), 12.72% to 14.05% (fb2, DP 25-36), and 10.80 to 20.72% (fb3, DP > 36), respectively. The contents of rapidly digestible starch (RDS), slowly digestible starch (SDS), and resistant starch (RS) ranged from 78.5% to 87.5%, 1.2% to 6.0%, and 10.1% to 18.0%, respectively. AAC was negatively correlated with RDS content but positively correlated with RS content in rice starch. The proportion of short chains in amylopectin, i.e. the amount of fraction IIa (FrIIa) fractionated by gel permeation chromatography (GPC), was positively correlated with RDS. Starch synthase IIa (SSIIa) gene controlled the degree of crystallinity, the amount of fa chains of amylopectin. SSIIIa gene controlled the amount of fb1 chains. Wx gene controlled the FrI, FrIIa, RDS, and RS. Starch debranching enzyme isoamylase II (ISA2) gene also controlled the RDS, which may suggest that RDS was also affected by amylopectin structure, although no correlation between them was found. This study indicated that genetics (i.e., starch biosynthesis related genes) controlled the structural properties of starch, and both amylose content and amylopectin fine structure determined functional properties of rice starch (i.e., the digestion), each in a different way. Understanding the "genetics-structure-function" relationships in rice starches will assist plant breeders and food processors in developing new rice varieties and functional foods.

The Role of Indica Starch in the Mechanism of Formation of Fresh Rice Noodles

Abstract:Starch is the largest component in frequently consumed fresh rice noodles (FRN). The effects of morphology, relative crystallinity, molecular weight distribution and physicochemical (rheological, water hydration and gel texture) properties of starch from five rice cultivars with different amylose contents on the formation of FRN were investigated. FRN made from NR rice cultivar, used for commercial rice noodles making, showed excellent FRN texture. NR starch exhibited larger G ' and smaller tan delta values. The morphology of FRN from GM03 cultivar was closest to NR, but had higher hardness and lower springiness, resilience and cohesiveness. Its starch granules morphology was relatively small and round. The extracted starch from Zhongzao 35, Zao 87 and BX02 rice cultivars, with higher swelling power or lower proportions of amylose (FrI) and higher proportions of amylopectin (FrII), could not produce a dense and consistent FRN structure. It was observed that relative crystallinity of the starch samples had no influence on the formation of FRN. Nevertheless, the gel texture of starch played a role in the formation of FRN. The formation of FRN is the common effect of structure and physicochemical properties of rice starch. The finding of this research would be useful in understanding the role of starch in producing FRN of high quality.

Abstract:Starch from 15 different rice genotypes with amylose content (AC) ranging 1.5%-30.6% were investigated for relationships between structures and properties. For parameters related to the granular level, the most important relationships were found for AC, average chain lengths (ACL) of the amylopectin (AP) fb1 chains having a length of DP 13-24, crystallinity, and the thickness of the crystalline (dc) and the amorphous lamellae (da) of the starch granule. AC and dc were negatively correlated with the peak gelatinization temperature (Tp), thermal enthalpy (ΔH), and peak viscosity (PV), but positively correlated with swelling power. ACLfb1 and da, as compared to AC and dc, had the opposite effects on these parameters, demonstrating important roles of specific molecular and lamellar structures on the starch granular stability. For the gelatinized systems, increasing ACLfb1 decreased retrogradation, while AC increased retrogradation by increasing the resistant starch (RS) content, storage modulus (G'), and setback (SB).

Abstract:Starch is the main source of dietary energy for humans. In order to understand the mechanisms governing native starch in vitro digestion, digestion data for six starches [wheat, maize, (waxy) maize, rice, potato and pea] of different botanical sources were fitted with the most common first-order kinetic models, i.e. the single, sequential, parallel and combined models. Parallel and combined models provided the most accurate fits and showed that all starches studied except potato starch followed a biphasic in vitro digestion pattern. The biological relevance of the kinetic parameters was explored by determining changes in crystallinity and molecular structure of the undigested starch residues during in vitro digestion. While the crystallinity of the undigested potato starch residues did not change substantially, a respectively small and large decrease in their amylose content and chain length during in vitro digestion was observed, indicating that amylose was digested slightly preferentially over amylopectin in native starch. However, the molecular structure of the starch residues changed too slowly and/or only to an insufficient extent to relate it to the kinetic parameters of the digested fractions predicted by the models. Such parameters thus need to be interpreted with caution, as their biological relevance still needs to be proven.
Correlation between in vitro starch digestibility and starch structure/physicochemical properties in rice

Abstract:Natural resistant starch (RS) in rice provides human health benefits, and its concentration in rice is influenced by the structure and physicochemical properties of starch. The native starch structures and physicochemical properties of three rice varieties, QR, BR58, and BR50, and their relationships to in vitro digestibility were studied. The starch granules in all three varieties were irregular or polyhedral in shape. There were a few oval granules and a few pinhole structures in QR, no oval granules but a higher number of pinholes in BR58, and no oval granules and pinholes in BR50. QR is a low-amylose (13.8 %), low-RS (0.2 %) variety. BR58 is a low-amylose (15.3 %), high-RS (6.5 %) variety. BR50 is a high-amylose (26.7 %), high-RS (8.3 %) variety. All three starches exhibited typical A-type diffraction patterns. Starch molecular weight, chain length distribution, starch branching degree, pasting capabilities, and thermal properties differed considerably between the rice starches. The RS contents of the rice starch varieties were positively correlated with AAC, Mw/Mn, Mz/Mn, peak 3, B, P Time , and Tp and negatively correlated with Mn, peak 2, DB, PV, and BD, according to Pearson's correlation analysis. These findings may be helpful for the breeding and development of high-RS rice varieties.

Abstract:Porous starch has been created through hydrolysis by amyloglucosidase and α-amylase. However, little information is known about the precise evolution of multi-scale structures of starch during digestion. In this study, rice starch and potato starch, containing different crystalline structures, were hydrolyzed by amyloglucosidase and α-amylase for 20 and 60 min, respectively, and their resulting structural changes were examined. The digestion process caused significant degradation of the molecular structures of rice and potato starches. In addition, the alterations in the ordered structures varied between the two starches. Rice starch exhibited porous structures, thicker crystalline lamellae as determined by small-angle X-ray scattering, and enhanced thermostability after digestion using differential scanning calorimetry. For rice starch, the extent of crystalline structures was analyzed with an X-ray diffractometer; it was found to first increase after 20 min of digestion and then decrease after 60 min of digestion. In contrast, potato starch did not display porous structures but exhibited thicker crystalline lamellae and a reduction in ordered structures after digestion. These findings suggest that it is possible to intentionally modulate the multi-scale structures of starch by controlling the digestion time, thereby providing valuable insights for the manipulation of starch functionalities.
Rheological, Thermal, and in Vitro Starch Digestibility Properties of Oat Starch-Lipid Complexes

Abstract:The influence of oat lipids on the structural, thermal, rheological, and in vitro digestibility properties of oat starch under heat processing conditions was investigated. X-ray diffraction, fourier infrared spectroscopy, and differential scanning calorimetry revealed the formation of a V-shaped crystal structure between starch and lipid, resulting in enhanced orderliness and enthalpy. Oat lipids decreased the final viscosity and gel strength of oat starch while weakening the trend towards gel network formation. Additionally, oat lipids exhibited enhanced resistance to starch hydrolase, leading to elevated contents of slowly digestible starch and resistant starch. Consequently, this leads to an augmentation in the rate constants for the rapid digestion fraction (k1) and the slow digestion fraction (k2). When the lipid content reached 7.50 %, a significant increase of 42.20 % was observed in the maximum digestibility of slow digestion fraction (C infinity 2), while a notable decrease of 44.06 % was noted in the maximum digestibility of rapid digestion fraction (C infinity 1). The correlation analysis revealed that lipid content, final viscosity, and enthalpy exerted significant influences on in vitro starch digestion. These results demonstrate the substantial impact of lipid content on oat starch structure, subsequently affecting its thermal, rheological, and digestive properties.
Structural characteristics and in vitro starch digestibility of oil-modified cooked rice with varied addition manipulations

Abstract:Lipid has crucial applications in improving the quality of starchy products during heat processing. Herein, the influence of lipid modification and thermal treatment on the physicochemical properties and starch digestibility of cooked rice prepared with varied addition manipulations was investigated. Rice bran oil (RO) and medium chain triglyceride oil (MO) manipulations were performed either before (BC) or after cooking (AC). GC-MS was applied to determine the fatty acid profiles. Nutritional quality was analyzed by quantifying total phenolics, atherogenic, and thrombogenic indices. All complexes exhibited higher surface firmness, a soft core, and less adhesive. FTIR spectrum demonstrated that the guest component affected some of the dense structural attributes of V-amylose. The kinetic constant was in the range between 0.47 and 0.86 min-1 wherein before mode presented a higher value. The lowest glucose release was observed in the RO_BC sample, whereas the highest complexing index was observed in the RO_AC sample, indicating that the dense molecular configuration of complexes that could resist enzymatic digestion was more critical than the quantity of complex formation. Despite the damage caused by mass and heat transfer, physical barrier, intact granule forms, and strengthened dense structure were the central contributors affecting the digestion characteristics of lipid-starch complexes.
Evaluation of the Molecular Structural Parameters of Normal Rice Starch and Their Relationships with Its Thermal and Digestion Properties

Abstract:The molecular structural parameters of six normal rice starches with different amylose contents were investigated through their iodine absorption spectra and gel permeation chromatography of fully branched and debranched starches. The thermal and digestion properties of starches were also determined and their relationships with molecular structural parameters were analyzed. Results showed that the molecular structural parameters of maximum absorption wavelength, blue value (BV), optical density 620 nm/550 nm (OD 620/550), amylose, intermediate component, and amylopectin, including its short branch-chains, long branch-chains, and branching degree, had high correlation in different determining methods. The intermediate component of starch was significantly positively related to amylose and negatively related to amylopectin, and the amylopectin branching degree was significantly positively related to amylopectin content and negatively related to amylose content. The gelatinization temperatures and enthalpy of native starch were significantly positively related to BV, OD 620/550, and amylose content and negatively related to amylopectin short branch-chains. The gelatinization temperatures and enthalpy of retrograded starch were significantly negatively related to amylopectin branching degree. The digestions of gelatinized and retrograded starches were significantly negatively related to the BV, OD 620/550, amylose, and intermediate component and positively related to amylopectin and its short branch-chains and branching degree.

Abstract:Starch granule-associated proteins (SGAPs) including granule-surface proteins and granule-channel proteins in waxy, low- and high-amylose rice starch were extracted and identified. The in vitro digestibility of starch was investigated before and after the extraction of granule-channel proteins or total SGAPs. The results showed that 10 types of major differentially expressed proteins (DEPs) including 14-3-3-like protein and ribosomal protein were found among starches. In addition, the lack of only granule-channel proteins or total SGAPs led to significant and different changes in the levels of rapidly digestible starch, slowly digestible starch and resistant starch. Possible mechanisms are related to the accessibility of amylase into starch granules and structural properties of SGAPs. This study provides more information about DEPs in rice starch with different amylose content and supports further study on the relationship between SGAPs and in vitro starch digestibility.
Structural changes of rice starch-anthocyanins complexes (V-type) and its impact on gut microbiotas and potential metabolic pathways during in vitro fermentation

Abstract:This study explored the differences in the in vitro fermentation properties of rice starch (RS) and rice starch-anthocyanins complexes (RS-A). Structural characterization suggested that RS and RS-A complexes showed a V-type crystalline structure. The degree of order (DO) and degree of double helix (DD) values of RS and RS-A complexes were enhanced after fermentation. Moreover, the RS-A complexes could improve the relative abundance of Bacteroidetes, Ruminococcaceae, and up-regulate gut microbiota diversity to maintain gut homeostasis. Relative abundance of potential metabolic pathways, such as energy metabolism, digestion system, and carbohydrate degradation overexpressed in the presence of RS-A complexes. The results demonstrated that the RS-A complexes had slower fermentation rates contributing to the transport of the formed short-chain fatty acid (SCFA) to the end of the colon and that the crystallinity might be a factor influencing the utilization of the starch matrix by the gut microbiota for SCFA formation.
Insights into the Multi-Scale Structure and in Vitro Digestibility Changes of Rice Starch-Oleic Acid/linoleic Acid Complex Induced by Heat-Moisture Treatment

Abstract:The study presents an innovative approach using heat-moisture treatment (HMT) to prepare rice starch-oleic acid (OA)/linoleic acid (LOA) complex. Relationship of the multi-scale structure-in vitro digestibility of rice starch-OA/LOA complex induced by HMT was established in this work. Herein, HMT induced the formation of single helices complex between OA/LOA and amylose or long side chains of amylopectin, which formed more V-type crystallite. HMT assisted complexation with OA/LOA resulted in higher ordered molecular chain aggregations in amorphous amylose background area, reduced the amorphous lamellae and made the double helices and A-type crystallite perfectly arranged. These structural alternations inhibited the molecular interactions (e.g., binding ability, catalyzed location) between rice starch and α-amylase, which eventually increased the resistant starch (RS) content and decreased the slow digestible starch (SDS) content. Moreover, HMT treated rice starch formed type II complex with OA and type I complex with LOA, and all the referred changes went more obvious with the increasing amount of OA than LOA. The results suggested that the OA/LOA was inserted inside the amylose spiral cavity and the amorphous lamellae of rice starch. The current study exhibited an in-depth research of the multi-scale structure within rice starch-OA/LOA complex and proposes a promising approach to ensure the starch products have designated digestibility.

Abstract:To investigate the multi-scale structural changes and digestibility of parboiled rice, the side chain distribution, helical structure, short/long-range ordered structure, and lamellar structure were systematically characterized and an in vivo postprandial blood glucose test was applied. The results indicate that parboiling has little effect on the side chain distribution, double helix content and helical structure order of parboiled rice. The crystal type of rice starch changed from type A to A + V or B + V after parboiling and the relative crystallinity decreased from 30.45 % to a minimum of 6.87 %. The in vivo study also indicated that parboiling significantly reduces the glycaemic index of rice to medium level. Our work is the first to focus on the parboiling conditions, multi-scale structural changes and in vivo digestibility of parboiled rice, which might provide guidance for the design of less digestible parboiled rice in the future.

In particular, milling reduced the content of saponins from 5.06% to 0.23%. Milling increased the swelling power of quinoa flour but decreased the gelatinization enthalpy of quinoa starch. Accordingly, the microstructure of quinoa with higher milling degree after cooking was more uniform and denser. Fourier transform infrared (FTIR) analysis showed that starch-protein interaction was beneficial to protect the ordered structure of quinoa starch. The digestibility of protein and starch was interrelated, and the digestibility rate increased with the raising milling degree. By fitting the starch digestograms and combining the digestion kinetic model, three digestion patterns of quinoa starch during milling were determined. The results of the confocal laser scanning microscope showed that the physical barrier composed of protein matrix coated starch and starch-protein complex affected starch digestion.

Abstract:The underlying mechanisms behind the genetics, structure and functionality relations in starches from various origins have not been fully understood. For better control of the genotypic background, rice starches from four chalky mutants and their parent were employed to investigate the above relations, and some interesting relations were revealed. GBSSI and SSIIIa were shown to affect the height of amylose in the debranched starch fraction (hAM) which reflects true amylose content (TAC), and then hAM was correlated with the AAC, RS, HD, COH, To, ΔHg, ΔHr, and R%. GBSSI also affected the average chain length (<math>X)¯</math> of amylopectin, which was associated with the AAC, TAC, RS, HPV, HD, COH, ΔHg, ΔHr, and R%. The SSI, SSIIa and SSIIIa affected the amylose size (Rh,AM), which was correlated with the TAC, AAC, RS, HD, COH, To and Tp. Furthermore, both SSIIa and PUL affected the XAP2, and XAP2 was correlated with the To.

Abstract:In this paper, we prepared a series of rice starch complexed with various konjac glucomannan (KGM) contents under screw extrusion. The complex's multi-scale structures, rheological and thermal properties, and in vitro digestion were explored. The complex's effects on short-chain fatty acids production and gut microbiota composition were further investigated using in vitro human fecal fermentation. Multi-scale structural analysis indicated that short-ordered degree, single/double helix structure contents, and crystallinity of the rice starch increased with an increased concentration of KGM. Rheology and differential scanning calorimeter analysis indicated that KGM enhanced the viscoelasticity of rice starch paste and the thermal stability of rice starch. In vitro digestion data revealed that as KGM concentrations increased, both slowly digested component (SDC) and resistant component (RC) contents increased. This phenomenon was ascribed to the complexation of KGM with rice starch assisted by screw extrusion. Further, the high RC content of the complex contributed to the yield of butyrate level, corresponding to the growth in Actinobacteria at the phylum level, and Megasphaera and Bifidobacterium at the genus level in the cultures. Overall, this research will be critical in determining the physicochemical properties and fecal fermentation behaviors of the rice starch–KGM complex and creating starchy foods with specific health benefits.
