Abstract:Herein the density functional theory (DFT) method was employed to investigate the reaction mechanism of enantioselective Cacchi reaction for N- aryl(alkyl)sulfonyl-2-alkynylanilides and PhB(OH)2 to form 2,3-disubstituted indoles catalyzed by palladium complex. The whole reaction mechanism includes two processes: the formation process of Pd-Ph species A6 and the cross-coupling process regarding the CN coupling within the substrate molecule and the CC coupling of -Ph and the substrate 1. The calculated results show that bases play an important role in the generation of Pd-Ph active species A6, and the rate-determining step is the CC coupling step with a free energy barrier of 18.0 kcal/mol in the formation of 2, 3-disubstituted indole. The enantioselectivity of the reaction is originated from the π-π stacking interaction, which is present in the naphthalene ring of the substrate and the benzene ring. The decomposition analysis and the reduced density gradient (RDG) analysis indicated that the enantioselectivity was mainly controlled by the combination of geometric deformation and non-covalent interaction (NCI) between the substrate and catalyst. This work could provide theoretical guidance for the synthesis of indoles with a chiral C2-aryl axis.
