Abstract:The synthesis of trifluoroethylamines as amide bond mimics is an interesting topic in current research. They are well known tools in pharmaceutical and agrochemical industry. Other methods described in literature are often restricted to few substrates. We herein report a new synthetic approach towards this class of substances. First, the trimethyl-aluminium-promoted generation of a trifluoromethyl-ketimine from the corresponding trifluoromethyl ketone and an aniline derivative was investigated. Next, the ketimine was converted into the trifluoroethylamines in a one-pot reaction by simple addition of borane-methyl sulfide complex.

Abstract:Enantioselective trifluoromethylation of aromatic ketones promoted by the cinchona alkaloid-derived ammonium bromide and sodium hydride was described. A series of trifluoromethyl-substituted aryl alcohols could be obtained in up to 82% ee with 98% yield under mild conditions. A possible catalytic cycle was also presented.

Abstract:Here we report a mild Ag-catalyzed domino acetalization reaction using quinoline-2-carboxylates, which were substructures in a number of natural products. Methanol was directly utilized as the source of the acetal part. The main features of the domino reaction include the Minisci reaction, oxidation, and acetalization. Hydrogen fluoride generated in-situ participates in the Minisci reaction, and no external acids were needed. This domino reaction provides a simple and versatile route to heteroaromatic acetals under mild condition with good functional group tolerance using methanol. (C) 2021 Elsevier Ltd. Abstract:A novel tandem reductive amination/intermolecular nucleophilic aromatic substitution (SNAr) sequence has been established for the synthesis of amine containing pyrimidine in formation of one carbon-oxygen and one carbon-nitrogen bonds in a one-pot fashion. Treatment of aldehyde with arylamine, 2-methanesulfonyl-4,6-dimethoxypyrimidine and sodium borohydride provides good overall yield. The p-toluenesulfonic acid (PTSA) can be used as activator and is generally needed in the reaction. Dioxane is the preferred reaction solvent, but reactions can also be carried out in tetrahydrofuran (THF), MeCN, toluene and dichloromethane. The procedure is carried out effectively in the presence of K2CO3. The reaction proceeds smoothly with aromatic aldehydes and arylamines possessing electron-donating or -withdrawing groups. This method can be applied to the synthesis of the oilseed rape herbicide and is superior to the classical one in several aspects: cutting out several purification steps, minimizing solvent use and chemical waste, and saving time. Its advantages such as operational convenience, high-efficient synthesis, and starting material availability make it a desirable method for preparing amines with molecular diversity and biological activity.

Abstract:Herein, we disclose a direct and powerful strategy for the synthesis of highly valuable α-trifluoromethylamine and N-trifluoroethylamine derivatives from a visible-light-promoted C,N-selective heteroarylation of N-trifluoroethyl hydroxylamine reagents with quinoxalin-2(1H)-ones under ambient conditions. The chemoselectivity of the process (trifluoroalkylation or N-trifluoroethylamination) can easily be dictated and modulated by a selection of N-trifluoroethyl hydroxylamine substrates. The key to success is the protecting group on the N atom of hydroxylamine reagents, which can control the process of 1,2-H shift of the in situ-generated N-trifluoroethyl radical. Remarkable features of this method include mild conditions, easy operation, high selectivity, and excellent functional group tolerability. More importantly, the trifluoroalkylated products can be readily derivatized into other interesting imidazo-fused heterocycles that would be of great potential for the exploitation of pharmaceutically relevant molecules.

Abstract:Amines are a fundamentally important class of biologically active compounds and the ability to manipulate their physicochemical properties through the introduction of fluorine is of paramount importance in medicinal chemistry. Current synthesis methods for the construction of fluorinated amines rely on air and moisture sensitive reagents that require special handling or harsh reductants that limit functionality. Here we report practical, catalyst-free, reductive trifluoroethylation reactions of free amines exhibiting remarkable functional group tolerance. The reactions proceed in conventional glassware without rigorous exclusion of either moisture or oxygen, and use trifluoroacetic acid as a stable and inexpensive fluorine source. The new methods provide access to a wide range of medicinally relevant functionalized tertiary β-fluoroalkylamine cores, either through direct trifluoroethylation of secondary amines or via a three-component coupling of primary amines, aldehydes and trifluoroacetic acid. A reduction of in situ-generated silyl ester species is proposed to account for the reductive selectivity observed.

Abstract:A method for catalytic regio- and enantioselective synthesis of trifluoromethyl-substituted and aryl-, heteroaryl-, alkenyl-, and alkynyl-substituted homoallylic a-tertiary NH2-amines is introduced. Easy-to-synthesize and robust N-silyl ketimines are converted to NH-ketimines in situ, which then react with a Z-allyl boronate. Transformations are promoted by a readily accessible L-threonine-derived aminophenol-based boryl catalyst, affording the desired products in up to 91% yield, >98:2 a:g selectivity, >98:2 Z:E selectivity, and >99:1 enantiomeric ratio. A commercially available aminophenol may be used, and allyl boronates, which may contain an alkyl-, a chloro-, or a bromo-substituted Z-alkene, can either be purchased or prepared by catalytic stereoretentive cross-metathesis. What is more, Z-trisubstituted allyl boronates may be used. Various chemo-, regio-, and diastereoselective transformations of the a-tertiary homoallylic NH2-amine products highlight the utility of the approach; this includes diastereo- and regioselective epoxide formation/trichloroacetic acid cleavage to generate differentiated diol derivatives.

Abstract:Herein, we report an example of a stereoselective γ-addition reaction of trifluoromethyl ketimines to 1-alkynyl ketones mediated by an isothiourea, BTM, under mild conditions, which afforded tetrasubstituted allenes with central chiralities in high yields (up to 94% yield), good enantioselectivities (up to 91% ee), and excellent diastereoselectivities (all >20 : 1 dr). In addition, the BTM-catalyzed γ-addition reaction was successfully applied to the gram-scale reaction, and an unexpected benzopyrrolothiazine derivative was successfully converted, albeit racemic.

Abstract:N-CF3 compounds constitute valuable targets in medicinal chemistry. Extensive studies have been reported for the preparation of N-CF3 compounds through fluorination and trifluoromethylation of N-containing compounds. The development of new synthetic methods from abundant and easily available substrates is highly desirable but still challenging. Herein, we report the design and synthesis of novel N-Cbz- and N-Boc-N-trifluoromethyl hydroxylamine reagents by silver-mediated oxidative trifluoromethylation. These reagents have been successfully applied to the direct incorporation of a NCF3 moiety into the commonly used unsaturated substrates under photoredox catalysis. This protocol enables the efficient and regioselective C-H trifluoromethylamination of various (hetero)arenes, including complex bioactive molecules. Furthermore, a variety of alkenes, dienes, and isonitriles undergo tandem trifluoromethylamination/functionalization delivering structurally diverse N-trifluoromethyl aliphatic and heteroaromatic amines. Notably, previously unknown cyclic N-CF3 compounds including N-CF3 oxazolidinones and oxazolones were conveniently prepared with N-Boc-N-trifluoromethyl hydroxylamine reagents. Furthermore, diversification of the resulting α-trifluoromethylamino ketones afforded the largely underexplored N-alkenyl- and N-alkynyl-N-CF3 compounds.

Abstract:The catalytic deoxyamination of readily available 2-arylethanols offers an appealing, simple, and straightforward means of accessing β-(hetero)arylethylamines of biological interest. Yet, it currently represents a great challenge to synthetic chemistry. In most cases, the alcohol has to be either pre-activated in situ or converted into a reactive carbonyl intermediate, limiting the substrate scope for some methods. Examples of direct dehydrative amination of 2-arylethanols are still scarce. Here, we describe a catalytic protocol based on the synergy of triflic acid and hexafluoroisopropanol, which enables the direct and stereospecific amination of a broad array of 2-arylethanols, and does not require any pre-activation of the alcohol. This approach yields high value-added products incorporating sulfonamide, amide, urea, and aniline functionalities. In addition, this approach was applied to the sulfidation of 2-arylethanols. Mechanistic experiments and DFT computations indicate the formation of phenonium ions as key intermediates in the reaction.

Abstract:In this work, a series of 4-aryl-3-(trifluoromethyl)-1 H -1,2,4-triazol-5(4 H )-one derivatives is synthesized through nucleophilic intramolecular cyclization reactions of ethyl 2-(2,2,2-trifluoro-1-(arylimino)ethyl)hydrazine-1-carboxylate intermediates, which are themselves prepared in high yields via reactions of ethyl chloroformate and N -aryl-2,2,2-trifluoroacetimidoyl chloride derivatives. Some of the merits of the reported procedure are an operationally simple and concise method, the ready availability of starting materials, excellent product yields, no formation of harmful by-products and easy purification of the products. Abstract:The asymmetric γ‐regioselective‐alkylation reaction which is catalyzed by a chiral tertiary amine ((DHQ)2PHAL) and takes advantage of isatin‐derived Morita‐Baylis‐Hillman carbonates and CF3‐contained ketoimines, delivered chiral α‐trifluoromethylamine derivatives and incorporated an oxindole motif in yields of 65‐98%, diastereoselectivities (> 20:1 in most cases) and enantioselectivities of 73‐99%. The subsequent deprotection gave α‐ trifluoromethyl amines in a stereoselective manner. Furthermore, fused polycyclic frameworks can be furnished by subsequent derivatization of the product without eroding its stereoselectivity. Abstract:As an alternative metal-free amination, we report a simple and efficient annulation reaction of 1-(3,3,3-trifluoro-2-oxopropyl)pyridin-1-ium bromide and α,β-unsaturated carbonyl compounds with NH4OAc or amines. With the developed protocol, a series of 3-trifluoromethyl aniline derivatives as the major products were obtained in good to excellent yields under Kröhnke pyridine synthesis conditions. The reaction proceeds via a cascade process involving the 1,4-Michael addition of the pyridinium ylide to α,β-unsaturated carbonyl compound, followed by intramolecular addition of carbanion to the keto carbonyl group to form a dienone intermediate.

Abstract:We have presented a straightforward approach for the synthesis of N-CF3 hydroxylamines, utilizing TMSCF3 for the N-trifluoromethylation of nitrosoarenes with the assistance of CsF. This method demonstrates excellent tolerance towards various functional groups and can be applied to a wide range of substrates, highlighting its versatility and utility in synthetic applications. The nucleophilic N-trifluoromethylation pathway for N-CF3 hydroxylamines provides a practical protocol for the synthesis of promising molecular architectures in the field of medicinal chemistry.

Abstract:A one-pot eco-friendly oxidative N -α-C(sp 3 )– H bond functionalisation of arylmethylamines for the synthesis of tetrasubstituted imidazoles is demonstrated. The substrate scope of these amines has been well-explored with different substrates, such as 1,2-diketones, an α-hydroxy ketone and phenylacetophenone. In the presence of FeCl 3 catalyst and green oxidant O 2 , the easily accessible substrates afforded tetrasubstituted imidazoles in up to 94% yield. Abstract:The rearrangement and demethylaromatization of cyclohexadienimines (namely cyclohexadienone imines) were detailedly investigated under metal-free conditions. Treating 4-aryl-4-methylcyclohexadienimines with acyl chloride at 100 °C in dichloromethane led to the smooth formation of m-arylaniline derivatives in good to excellent yields, in which [1,2]-migration of aryl group at C-4 occurred exclusively. The demethylaromatization of 4-aryl-4-methylcyclohexadienimines mediated by iodotriphenylphosphonium iodide (in-situ prepared via the reaction of triphenylphosphine with iodine) in toluene at 100 °Cproceeded well generating p-arylanilines in moderate to good yields. An efficient and alternate method for the synthesis of polysubstituted aryl amines especially m-arylaniline derivatives which are otherwise difficult to synthesize through traditional method was developed.
