Abstract:A phosgene- and metal-free synthesis of unsymmetrical arylurea derivatives utilizing 3-substituted dioxazolones and commercially available amines has been developed. The 3-substituted dioxazolones serve as the precursors of thein situgenerated isocyanate intermediates in the presence of non-toxic sodium acetate as a base and methanol as a solvent under mild heating conditions. A series of unsymmetrical phenylureas includingN, N'- mono-, di-, and trisubstituted derivatives are chemoselectively obtained in moderate to excellent yields from a broad scope of both 3-substituted dioxazolones and amines without a significant amount of the symmetrical byproducts. In many cases, the resulting unsymmetrical phenylureas are easily separated by filtration allowing no chromatographic purification. These investigations provide an opportunity for facile, practical, and eco-friendly synthesis of unsymmetrical diarylureas. Application of this method is demonstrated toward the gram-scale preparation of anti-cancer drugs, sorafenib and the 3-cycle continuous synthesis in conjugation with the recycle of sodium acetate and methanol process of known herbicide, daimuron.

Abstract:A new practical approach to prepare 1,3-(diethoxymethyl)-5-N,N-dimethylamino-6-methyluracil (5) has been realized from 6-methyluracil (1). Compound 5 is a key intermediate material in the synthesis of HIV-1 RT and methionine synthase inhibitors. 6-Methyluracil (1) reacted with HNO3-P2O5 giving 5-nitro-6-methyluracil (2), which alkylated at N-1,N-3 through reaction with ethoxymethyl chloride to give 3. Compound 3 was then reduced through treatment with HCOONH4-Pd/C and the derived C-5 amino group reacted with CH3I to convert into compound 5 in good yield.

Abstract:The reactions of α-bromoacetophenones with methylhydrazine in refluxing acetic acid generated 2-methyl-4-aryl-2H-[1,2,3]triazoles in good yields. The method was developed by the reactions of α-bromoacetophenones with phenylhydrazines in the presence of cupric ion, leading to 2,4-diary-2H-[1,2,3]triazoles. The structures were established on the basis of corresponding IR, 1H NMR, and elemental analysis data.

Abstract:The development of a new and straightforward chemoselective method for the synthesis of uracil-based structures by combining Suzuki–Miyaura and Sonogashira–Hagihara cross-coupling is reported. The methodology was applied to synthesize a series of novel compounds. The tolerance of the combination of different functional groups was tested. The influence of different functional groups on the physical properties was studied by ultraviolet–visible (UV–vis) and fluorescence spectroscopy, providing new insights into the potential applications of uracil-based structures.

Abstract:A novel and convenient one-step method for the synthesis of 1,3,4,-Triaryl-substituted-3,4,5,6-tetrahydropyrimidin-2(1H)-ones (3) was developed by cyclization of aromatic isocyanates (1) with 3-arylamino-1-phenylpropan-1-ones (2) under refluxing of toluene. Effects of reaction conditions on the converting ratio and yield were investigated. The method offered several advantages including simple experimental procedure, easily available starting materials and excellent yields (79%similar to 96%).

Abstract:A stereoselective radical-mediated hydrogermylation of the protected 5-ethynyluracil nucleosides with trialkyl-, triaryl,- or tris(trimethylsilyl)germanes gave (Z)-5-(2-germylvinyl)uridine, 2'-deoxyuridine, or ara-uridine as major products. Reaction of the beta-triphenylgermyl vinyl radical intermediate with oxygen and fragmentation of the resulting peroxyradical provided also 5-[2-(triphenylgermyl)acetyl]pyrimidine nucleosides in low to moderate yields. Thermal isomerization of the latter in MeOH occurred via a four-centered activated complex, and subsequent hydrolysis of the resulting O-germyl substituted enol yielded 5-acetyluracil nucleosides in quantitative yield.

Abstract:We have developed a novel and efficient method for the preparation of 2,4-disubstituted-1,2,3-triazoles. These compounds 3a-3p were synthesized in good yield by refluxing alpha-aminoacetophenones and hydrazines in glacial acetic acid. The reaction proceeds under mild conditions and is compatible with various functional groups.

Abstract:3-Alkyl-5-(diethylaminomethyl)pyrazoles reacted with chloroacetyl chloride to give 1-chloroacetyl derivatives which were subjected to heterocyclization with thiobenzamide, thiourea, and N -phenylthiourea in the presence of triethylamine in ethanol. As a result, previously unknown 3-alkyl-5-(diethylaminomethyl)-1-(1,3-thiazol-4-yl)-1 H -pyrazoles were synthesized.

Abstract:The regioselective reaction of 4,5-dihydroxy-1-methylimidazolidine-2-thione with 1-alkylureas was explored for the first time, predominantly providing the previously inaccessible 1-alkyl-4-methylsemithioglycolurils as the major products. We demonstrated that the interaction of the reactants was accompanied by the formation of isomeric 1-alkyl-6-methylsemithioglycolurils and several known compounds: 1-methyl- and 3-methylthiohydantoins, 1-methyl-1 H -imidazole-2(3 H )-thione, and 1,4-disubstituted glycolurils as byproducts. The reaction of 4,5-dihydroxy-1-methylimidazolidine-2-thione with 1-isopropylurea also produced a mixture of 1,1'-(1-methyl-2-thioxoimidazolidine-4,5-diyl)bis(3-isopropylurea) diastereomers.

Unveiling the Synthesis, Spectral Characterizations, and Electrochemical Potential of Novel (E)-Furan-2-yl Acrylohydrazides: An Exploration in Molecular Design

Abstract:In this study, we present the synthesis of novel derivatives of 3-furan-2-yl acrylohydrazide using a meticulous three-step reaction sequence. The synthesis ends up in the condensation of (E)-3-(furan-2-yl) acrylohydrazine (3) with diverse benzaldehyde and acetophenone derivatives. Comprehensive characterization of the synthesized compounds was achieved through 1D NMR spectroscopic analyses (1H and 13C NMR), 2D NMR spectroscopy (HSQC, NOESY), and high-resolution mass spectrometry (HRMS). The investigation of 1H NMR data at room temperature in deuterated dimethyl sulfoxide (DMSO-d6) unveiled the existence of (E)-3-(furan-2-yl) acrylohydrazide derivatives (4a-h) in a conformational equilibrium, manifesting as a mixture of synperiplanar E (sp E) and antiperiplanar E (ap E), or synperiplanar Z (sp Z) and antiperiplanar Z (ap Z). Notably, compounds 4a and 4b predominantly adopted the sp E conformer (Ec=c sp EC=N), while compounds 4c and 4d favored the antiperiplanar conformation. For the remaining compounds (4e-h), both conformers were nearly equimolar, with a marginal preference for the anti over the syn conformer. Interestingly, compounds 4f and 4h exhibited a prevalence of the apZ conformer (Ec=c ap ZC=N), while compound 4e featured the apE conformer (Ec=c ap EC=N). UV-visible absorption spectra for the N-acylhydrazones (4a-h) indicated absorption within the 570-635 nm range. Furthermore, cyclic voltammetry results demonstrated the capacity of the synthesized (E)-3-(furan-2-yl) acrylohydrazide derivatives (4a-h) to undergo quasi-reversible oxidation and reduction processes on a platinum electrode. These findings contribute valuable insights into the conformational dynamics and electrochemical behavior of this class of compounds, holding significance for applications in diverse scientific and technological domains.

Abstract:The reaction of thioxopyrimidine derivative 1a with chloroacetic acid and p-chlorobenzaldehyde gave compound 2. Alkylation of 1b using chloroacetonitrile gave N-alkylated derivative 6 which was reacted with hydrazine hydrate to give 7. Compound 7 was cyclized with formic and nitrous acid to give 8 and 9, respectively. The reaction of 1b with phosphorous oxychloride gave chloropyrimidine derivative 10 which was reacted and cyclized with different reagents to give compounds 11–23. The antimicrobial activity was studied against examples of Gram-positive and Gram-negative bacteria.

Abstract:Synthesis of 3-acy1-5,6-dihydro-1,4-oxathiines has been achieved via reactions of thiiranes and alpha-diazo-beta-1,3-dicarbonyl compounds under microwave and copper sulfate-assisted conditions. The current method provides a direct and simple strategy in efficient preparation of 3-acy1-1,4-oxathiines from readily available thiiranes and trans-3-acy1-5,6-dihydro-1,4-oxathiines as stereospecific products for 1,2-disubstituted cis-thiiranes. Abstract:The deprotection of 1,3-dibenzyl-5-(N,N-dimethylamino)-6-ethyluracil 1 was investigated. A practical, regioselective N-3 deprotection of compound 1 was performed with excellent yield using cyclohexene as a hydrogen donor.

Abstract:A mechanochemistry approach is developed for regioselective synthesis of functionalized dihydropyrido[2,3‐d]pyrimidines by milling propargylic alcohols and 6‐amino uracils with HFIP/p‐TsOH. In the case of tert‐propargyl alcohols, this [3+3] cascade annulation proceeded through allenylation of uracil followed by a 6‐endo trig cyclization. With sec‐propargyl alcohols, the reaction furnished the propargylation of uracil. This atom economy ball milling reaction allows access to a broad range of dihydropyrido[2,3‐d]pyrimidine derivatives in excellent yields. We demonstrated the gram scale synthesis of 3g and post‐synthetic modifications to effect the cyclization of 5 to 6.
