Abstract:Sufentanil in combination with dezocine or esketamine is often used for postoperative analgesia. However, there is a lack of clinical evidence of efficacy. This study compares the analgesic effects of esketamine and dezocine combined with sufentanil for relieving pain after laparoscopic cholecystectomy(LC).

However, when used alone, it may cause adverse reactions such as respiratory depression, nausea and vomiting. Therefore, it is usually combined with other analgesic drugs to enhance the analgesic effect and reduce adverse reactions. 3. **Dezocine and Esketamine**: - Dezocine is a partial agonist, which exerts analgesic and sedative effects by inhibiting the re - absorption of norepinephrine and serotonin. - Esketamine is an N - methyl - D - aspartic acid (NMDA) receptor antagonist, which has the advantages of high bioavailability, rapid onset and metabolism, and few side effects. It has been widely used in cancer pain control, antidepressant treatment and peri - operative analgesia. ### Research Objectives 1. **Compare Analgesic Effects**: Evaluate the visual analogue scale (VAS) scores of esketamine combined with sufentanil and dezocine combined with sufentanil at 4 hours, 8 hours, 24 hours and 48 hours after the operation. 2. **Evaluate Inflammatory Factor Levels**: Measure the levels of interleukin - 6 (IL - 6) and C - reactive protein (CRP) at 24 hours and 48 hours after the operation to evaluate the effects of the two groups of drugs on the inflammatory response. 3. **Evaluate Sleep Quality**: Use the Athens Insomnia Scale (AIS) to evaluate the sleep quality of patients one month before the operation, one day and two days after the operation. 4. **Monitor Adverse Reactions**: Record the incidence of postoperative nausea, vomiting, dizziness, headache and other adverse reactions in the two groups of patients. ### Main Findings 1. **Analgesic Effects**: The VAS scores of the esketamine combined with sufentanil group at 4 hours, 8 hours, 24 hours and 48 hours after the operation were significantly lower than those of the dezocine combined with sufentanil group. 2. **Inflammatory Factor Levels**: The levels of IL - 6 and CRP in the esketamine combined with sufentanil group at 24 hours and 48 hours after the operation were significantly lower than those in the dezocine combined with sufentanil group. 3. **Sleep Quality**: The AIS scores of the esketamine combined with sufentanil group one day and two days after the operation were significantly lower than those of the dezocine combined with sufentanil group, indicating that esketamine is helpful to improve postoperative sleep quality. 4. **Adverse Reactions**: There was no significant difference in the incidence of adverse reactions between the two groups of patients, and the incidence of nausea and vomiting in the esketamine group did not increase significantly. ### Conclusion Esketamine combined with sufentanil is superior to dezocine combined with sufentanil in relieving pain after laparoscopic cholecystectomy, reducing the levels of inflammatory factors and improving postoperative sleep quality, and will not increase the incidence of adverse reactions. This finding provides a new postoperative analgesia scheme for the clinic.
