Abstract:Migratory songbirds have the remarkable ability to extract directional information from the Earth's magnetic field1,2. The exact mechanism of this light-dependent magnetic compass sense, however, is not fully understood. The most promising hypothesis focuses on the quantum spin dynamics of transient radical pairs formed in cryptochrome proteins in the retina3-5. Frustratingly, much of the supporting evidence for this theory is circumstantial, largely because of the extreme challenges posed by genetic modification of wild birds. Drosophila has therefore been recruited as a model organism, and several influential reports of cryptochrome-mediated magnetic field effects on fly behaviour have been widely interpreted as support for a radical pair-based mechanism in birds6-23. Here we report the results of an extensive study testing magnetic field effects on 97,658 flies moving in a two-arm maze and on 10,960 flies performing the spontaneous escape behaviour known as negative geotaxis. Under meticulously controlled conditions and with vast sample sizes, we have been unable to find evidence for magnetically sensitive behaviour in Drosophila. Moreover, after reassessment of the statistical approaches and sample sizes used in the studies that we tried to replicate, we suggest that many-if not all-of the original results were false positives. Our findings therefore cast considerable doubt on the existence of magnetic sensing in Drosophila and thus strongly suggest that night-migratory songbirds remain the organism of choice for elucidating the mechanism of light-dependent magnetoreception.

Abstract:The notion that animals can detect the Earth's magnetic field was once ridiculed, but is now well established. Yet the biological nature of such magnetosensing phenomenon remains unknown. Here, we report a putative magnetic receptor (Drosophila CG8198, here named MagR) and a multimeric magnetosensing rod-like protein complex, identified by theoretical postulation and genome-wide screening, and validated with cellular, biochemical, structural and biophysical methods. The magnetosensing complex consists of the identified putative magnetoreceptor and known magnetoreception-related photoreceptor cryptochromes (Cry), has the attributes of both Cry- and iron-based systems, and exhibits spontaneous alignment in magnetic fields, including that of the Earth. Such a protein complex may form the basis of magnetoreception in animals, and may lead to applications across multiple fields.
Microgravity simulation by diamagnetic levitation: effects of a strong gradient magnetic field on the transcriptional profile of Drosophila melanogaster

Abstract:BackgroundMany biological systems respond to the presence or absence of gravity. Since experiments performed in space are expensive and can only be undertaken infrequently, Earth-based simulation techniques are used to investigate the biological response to weightlessness. A high gradient magnetic field can be used to levitate a biological organism so that its net weight is zero.ResultsWe have used a superconducting magnet to assess the effect of diamagnetic levitation on the fruit fly D. melanogaster in levitation experiments that proceeded for up to 22 consecutive days. We have compared the results with those of similar experiments performed in another paradigm for microgravity simulation, the Random Positioning Machine (RPM). We observed a delay in the development of the fruit flies from embryo to adult. Microarray analysis indicated changes in overall gene expression of imagoes that developed from larvae under diamagnetic levitation, and also under simulated hypergravity conditions. Significant changes were observed in the expression of immune-, stress-, and temperature-response genes. For example, several heat shock proteins were affected. We also found that a strong magnetic field, of 16.5 Tesla, had a significant effect on the expression of these genes, independent of the effects associated with magnetically-induced levitation and hypergravity.ConclusionsDiamagnetic levitation can be used to simulate an altered effective gravity environment in which gene expression is tuned differentially in diverse Drosophila melanogaster populations including those of different age and gender. Exposure to the magnetic field per se induced similar, but weaker, changes in gene expression.

The Power of Drosophila Genetics in Studying Insect Toxicology and Chemical Ecology

Abstract:Insect toxicology and chemical ecology are inherently interconnected disciplines, both dedicated to unraveling the intricate relationships between insects and the diverse array of chemical compounds that pervade their surroundings. Drosophila melanogaster , owing to its genetic and physiological similarities to other insects, serves as a robust model system in the study of insect toxicology. Moreover, state-of-the-art techniques in Drosophila neurobiology have extensively probed the chemosensory system of insects, providing significant insights into their adaptation to chemical environments. In this review, we emphasize the advancements achieved through the application of Drosophila genetics in investigations spanning both of these fields, significantly enhancing our understanding of the mode of action and resistance mechanisms of insecticides, as well as unraveling the molecular and cellular mechanisms underlying insect chemosensation and associated behaviors. The profound insights derived through this tiny fly not only enrich our understanding of the broader world of insects but also hold the potential to develop more effective and sustainable strategies for pest management.
Exposure to hypomagnetic field space for multiple generations causes amnesia in Drosophila melanogaster.

Abstract:This is the introduction of Drosophila into the study of learning and memory affected by removal of the geomagnetic field (GMF) for successive generations. Using the operant visual learning/memory paradigm at a flight simulator, the present study revealed that wild-type flies raised in a hypomagnetic field environment continuously for 10 successive generations were gradually impaired in visual conditioning learning and memory formation and finally the 10th generation flies became morphs of nonlearners and completely amnesiac. The control experiments show that the impairment could not be ascribed to any apparent sensorimotor problems in Drosophila. The reverse shift from hypomagnetic field (HMF) to natural GMF restored the GMF-free induced amnesia fully after six consecutive generations. Thus, our findings demonstrate conclusively that some serious, but reversible learning and memory impairment may occur for living organisms in a prolonged separation from GMF over many consecutive generations. And Drosophila has the potential to develop into a new model organism for the study of the neurobiology of magnetism for multiple generations.
Sub-second multi-channel magnetic control of select neural circuits in behaving flies

Abstract:Abstract Precisely timed activation of genetically targeted cells is a powerful tool for studying neural circuits and controlling cell-based therapies. Magnetic control of cell activity or “magnetogenetics” using magnetic nanoparticle heating of temperature-sensitive ion channels enables remote, non-invasive activation of neurons for deep-tissue applications and studies of freely behaving animals. However, the in vivo response time of thermal magnetogenetics is currently tens of seconds, which prevents the precise temporal modulation of neural activity similar to light-based optogenetics. Moreover, magnetogenetics has not provided a means to selectively activate multiple channels to drive behavior. Here we produce sub-second behavioral responses in Drosophila melanogaster by combining magnetic nanoparticles with a rate-sensitive thermoreceptor (TRPA1-A). Furthermore, by tuning the properties of magnetic nanoparticles to respond to different magnetic field strengths and frequencies, we can achieve sub-second, multichannel stimulation, analogous to multi-color optogenetic stimulation. These results bring magnetogenetics closer to the temporal resolution and multiplexed stimulation possible with optogenetics while maintaining the minimal invasiveness and deep-tissue stimulation only possible by magnetic control.
Cryptochromes and Hormone Signal Transduction under Near-Zero Magnetic Fields: New Clues to Magnetic Field Effects in a Rice Planthopper

Abstract:Although there are considerable reports of magnetic field effects (MFE) on organisms, very little is known so far about the MFE-related signal transduction pathways. Here we establish a manipulative near-zero magnetic field (NZMF) to investigate the potential signal transduction pathways involved in MFE. We show that exposure of migratory white-backed planthopper, Sogatella furcifera, to the NZMF results in delayed egg and nymphal development, increased frequency of brachypterous females, and reduced longevity of macropterous female adults. To understand the changes in gene expression underlying these phenotypes, we examined the temporal patterns of gene expression of (i) CRY1 and CRY2 as putative magnetosensors, (ii) JHAMT, FAMeT and JHEH in the juvenile hormone pathway, (iii) CYP307A1 in the ecdysone pathway, and (iv) reproduction-related Vitellogenin (Vg). The significantly altered gene expression of CRY1 and CRY2 under the NZMF suggest their developmental stage-specific patterns and potential upstream location in magnetic response. Gene expression patterns of JHAMT, JHEH and CYP307A1 were consistent with the NZMF-triggered delay in nymphal development, higher proportion of brachypterous female adults, and the shortened longevity of macropterous female adults, which show feasible links between hormone signal transduction and phenotypic MFE. By conducting manipulative NZMF experiments, our study suggests an important role of the geomagnetic field (GMF) in modulating development and physiology of insects, provides new insights into the complexity of MFE-magnetosensitivity interactions, and represents an initial but crucial step forward in understanding the molecular basis of cryptochromes and hormone signal transduction involved in MFE.

Abstract:An atomic magnetometer (AM) was used to non-invasively detect the tiny magnetic field generated by the brain of a single Drosophila. Combined with a visual stimulus system, the AM was used to study the relationship between visual salience and oscillatory activity of the Drosophila brain by analyzing changes in the magnetic field. Oscillatory activity of Drosophila in the 1-20 Hz frequency band was measured with a sensitivity of 20 fT root Hz. The field in the 20-30 Hz band under periodic light stimulation was used to explore the correlation between short-term memory and visual salience. Our method opens a new path to a more flexible method for the investigation of brain activity in Drosophila and other small insects.
Application of Mechanical Forces on Drosophila Embryos by Manipulation of Microinjected Magnetic Particles

Abstract:Cells generate mechanical forces to shape tissues during morphogenesis. These forces can activate several biochemical pathways and trigger diverse cellular responses by mechano-sensation, such as differentiation, division, migration and apoptosis. Assessing the mechano-responses of cells in living organisms requires tools to apply controlled local forces within biological tissues. For this, we have set up a method to generate controlled forces on a magnetic particle embedded within a chosen tissue of Drosophila embryos. We designed a protocol to inject an individual particle in early embryos and to position it, using a permanent magnet, within the tissue of our choice. Controlled forces in the range of pico to nanonewtons can be applied on the particle with the use of an electromagnet that has been previously calibrated. The bead displacement and the epithelial deformation upon force application can be followed with live imaging and further analyzed using simple analysis tools. This method has been successfully used to identify changes in mechanics in the blastoderm before gastrulation. This protocol provides the details, (i) for injecting a magnetic particle in Drosophila embryos, (ii) for calibrating an electromagnet and (iii) to apply controlled forces in living tissues.
Weak magnetic field effects in biology are measurable—accelerated embryogenesis in the absence of the geomagnetic field

Abstract:Despite decades of reports of weak magnetic field effects in biology across the tree of life and on a broad range of cell types, the evidence to date remains met with skepticism. To remedy this, we present open-data, large scale, and varied morphological evidence that embryo development is accelerated in a well engineered, environmentally-calibrated hypomagnetic field of less than 1 nT. These data imply that basal tadpole physiology can sense and react to the absence of Earth's minute magnetic field of approximately 50 µT. The effect is significant, as demonstrated by a variety of statistical measures. As no definitive biophysical mechanism has been identified to account for its occurrence, this study raises the question of which mechanism provides the most plausible explanation. How that question is answered may have implications in a variety of fields, including human health, behavioral ecology, and space exploration.

Abstract:It is a well established notion that animals can detect the Earth's magnetic field, while the biophysical origin of such magnetoreception is still elusive. Recently, a magnetic receptor Drosophila CG8198 (MagR) with a rod-like protein complex is reported [Qin \emph{et al}., Nat. Mater. \textbf{15}, 217 (2016)] to act like a compass needle to guide the magnetic orientation of animals. This view, however, is challenged [Meister, Elife \textbf{5}, e17210 (2016)] by arguing that thermal fluctuations beat the Zeeman coupling of the proteins's magnetic moment with the rather weak geomagnetic field ($\sim25-65$ $\mu$T). In this work, we show that the spin-mechanical interaction at the atomic scale gives rise to a high blocking temperature which allows a good alignment of protein's magnetic moment with the Earth's magnetic field at room temperature. Our results provide a promising route to resolve the debate on the thermal behaviors of MagR, and may stimulate a broad interest on spin-mechanical couplings down to atomistic levels.
Modulation of the cardiac pacemaker of Drosophila: cellular mechanisms

Abstract:The myogenic cardiac pacemaker of Drosophila melanogaster responds to a range of neurotransmitters and hormones by adjusting heart rate. These cardioactive substances ultimately affect the activity of ion channels comprising the pacemaker. We report here work utilizing genetic variants and pharmacological tools to explore a subset of possible mechanisms for this cellular signaling, specifically: receptors, cAMP, cGMP, G proteins, and calcium. We found that alpha(1) adrenergic and 5-hydroxytryptamine(2) (5-HT(2)) receptors are critical components of mediating modulation of heart rate. There was no evidence that the cAMP system is part of the modulatory mechanism. cGMP is likely to be integral to one active pathway, as non-hydrolyzable forms of this cyclic nucleotide increase heart rate, and flies bearing the mutation sitter, a recessive allele of the foraging gene, which encodes a cGMP-dependent kinase, have tachycardia. Heart rhythm is affected by pertussis toxin and by agonists and antagonists of both alpha(1) adrenergic and 5-HT(2) receptors; this suggests involvement of two different types of G proteins. The l(4)16/ciD line, containing a mutation in CaM kinase II, eliminates pacemaker responsiveness to serotonin but is without effect on norepinephrine sensitivity. This result is the same as that for the CaM kinase II enzyme inhibitor KN-93. This work establishes a framework for further investigations into the control of the cardiac pacemaker, and expands the applicability of the Drosophila heart model.
Magnetic field platform for experiments on well-mixed and spatially structured microbial populations

Abstract:Magnetic fields have been shown to affect sensing, migration, and navigation in living organisms. However, the effects of magnetic fields on microorganisms largely remain to be elucidated. We develop an open-source, 3D-printed magnetic field exposure device to perform experiments on well-mixed and spatially structured microbial populations. This device is designed in AutoCAD, modeled in COMSOL, and validated using a Gaussmeter and experiments on the budding yeast Saccharomyces cerevisiae. We find that static magnetic field exposure slows the spatially structured expansion of yeast mats that expand in two dimensions, but not yeast mats that expand in three dimensions, across the surface of semi-solid yeast extract-peptone-dextrose agar media. We also find that magnetic fields do not affect the growth of planktonic yeast cells in well-mixed liquid yeast extract-peptone-dextrose media. This study provides an adaptable device for performing controlled magnetic field experiments on microbes and advances our understanding of the effects of magnetic fields on fungi.

Abstract:Significance Although migratory animals, such as salmon and sea turtles, are thought to imprint on their natal magnetic field and use this information to return as adults, direct experimental evidence thereof remains elusive. This study experimentally demonstrates that organisms can imprint a magnetic field and that this imprinting can be inherited by their progeny. Prenatal exposure to a specific geographic magnetic field during development affected adult responses to the matching field gradient through downward movements associated with foraging. This same behavior occurred spontaneously in the progeny of the next generation. These results suggest that imprinting on the magnetic field of a natal area assists magnetoreceptive organisms and their offspring in recognizing locations favorable for foraging and reproduction.

Abstract:This is an analysis of how magnetic fields affect biological molecules and cells. It was prompted by a series of prominent reports regarding magnetism in biological systems. The first claims to have identified a protein complex that acts like a compass needle to guide magnetic orientation in animals ( Qin et al., 2016 ). Two other articles report magnetic control of membrane conductance by attaching ferritin to an ion channel protein and then tugging the ferritin or heating it with a magnetic field ( Stanley et al., 2015 ; Wheeler et al., 2016 ). Here I argue that these claims conflict with basic laws of physics. The discrepancies are large: from 5 to 10 log units. If the reported phenomena do in fact occur, they must have causes entirely different from the ones proposed by the authors. The paramagnetic nature of protein complexes is found to seriously limit their utility for engineering magnetically sensitive cells.

Biological cell as a soft magnetoelectric material: Elucidating the physical mechanisms underpinning the detection of magnetic fields by animals

Abstract:Sharks, birds, bats, turtles, and many other animals can detect magnetic fields. Aside from using this remarkable ability to exploit the terrestrial magnetic field map to sense direction, a subset is also able to implement a version of the so-called geophysical positioning system. How do these animals detect magnetic fields? The answer to this rather deceptively simple question has proven to be quite elusive. The currently prevalent theories, while providing interesting insights, fall short of explaining several aspects of magnetoreception. For example, minute magnetic particles have been detected in magnetically sensitive animals. However, how is the detected magnetic field converted into electrical signals given any lack of experimental evidence for relevant electroreceptors? In principle, a magnetoelectric material is capable of converting magnetic signals into electricity (and vice versa). This property, however, is rare and restricted to a rather small set of exotic hard crystalline materials. Indeed, such elements have never been detected in the animals studied so far. In this work we quantitatively outline the conditions under which a biological cell may detect a magnetic field and convert it into electrical signals detectable by biological cells. Specifically, we prove the existence of an overlooked strain-mediated mechanism and show that most biological cells can act as nontrivial magnetoelectric materials provided that the magnetic permeability constant is only slightly more than that of a vacuum. The enhanced magnetic permeability is easily achieved by small amounts of magnetic particles that have been experimentally detected in magnetosensitive animals. Our proposed mechanism appears to explain most of the experimental observations related to the physical basis of magnetoreception.

Abstract:Increasing numbers of behavioral observations have shown that many organisms can sense magnetic fields in the environment. However, the underlying mechanisms of magnetosensation remain elusive, and a major problem is the lack of an animal model convenient for detecting magnetic responses, especially in vertebrates. We have previously reported that the physical performance of mice decreased after 1 month of exposure to a hypomagnetic field, which probably includes a light-independent mechanism. In this study, we investigated the effects of a static magnetic field on the locomotion of Xenopus tadpoles (stage 48) using a real-time recording system. Because it is speculated that the photosensing system is related to magnetosensation, we triggered the tadpoles to swim by a 30 s light pulse after a 5 min adaptation in the dark and measured the swimming activities of the tadpoles under static magnetic field exposure in both the dark and bright conditions. Compared to the sham control treatment, the presence of a magnet inhibited the movement of the tadpoles under both bright and dark conditions, as shown by reductions in swimming distance, speed, and counts of path adjustment. Moreover, the directional preference for path adaptation was altered when swimming in the dark. These results suggest that tadpoles in the dark can exert a rapid locomotion response to changes in the environmental magnetic field, providing evidence for an intrinsic, light-independent rapid magnetoresponse pathway in tadpoles. In addition, this rapid vertebrate locomotion assay paradigm will be a practical tool to facilitate further investigation of the mechanisms of biomagnetic effects.

Abstract:Abstract Many animals use the Earth’s geomagnetic field for orientation and navigation. Yet, the molecular and cellular underpinnings of the magnetic sense remain largely unknown. A biophysical model proposed that magnetoreception can be achieved through quantum effects of magnetically-sensitive radical pairs formed by the photoexcitation of cryptochrome (CRY) proteins. Studies in Drosophila are the only ones to date to have provided compelling evidence for the ultraviolet (UV)-A/blue light-sensitive type 1 CRY (CRY1) involvement in animal magnetoreception, and surprisingly extended this discovery to the light-insensitive mammalian-like type 2 CRYs (CRY2s) of both monarchs and humans. Here, we show that monarchs respond to a reversal of the inclination of the Earth’s magnetic field in an UV-A/blue light and CRY1, but not CRY2, dependent manner. We further demonstrate that both antennae and eyes, which express CRY1, are magnetosensory organs. Our work argues that only light-sensitive CRYs function in animal light-dependent inclination-based magnetic sensing.

Abstract:We report Static Magnetic Field (SMF) induced altered sub-cellular streaming, which retains even after withdrawal of the field. The observation is statistically validated by differential fluorescence recovery after photo-bleaching (FRAP) studies in presence and absence of SMF, recovery rate being higher in presence of SMF. This instant magneto-sensing by live cells can be explained by inherent diamagnetic susceptibility of cells and alternatively by spin recombination, e.g., by the radical pair mechanism. These arguments are however insufficient to explain the retention of the SMF effect even after field withdrawal. Typically, a relaxation time scale at least of the order of minutes is observed. This long duration of the SMF effect can be explained postulating a field induced coherence that is followed by decoherence after the field withdrawal. A related observation is the emergence of enhanced magnetic susceptibility of cells after magnetic pre-incubation. This implies onset of a new spin equilibrium state as a result of prolonged SMF incubation. Lastly, translation of such altered spin states to a cellular signal that leads to an altered sub-cellular streaming, probable intracellular machineries for this translation being discussed in the text.

Abstract:Unlike mammals, insects such as Drosophila have multiple taste organs. The chemosensory neurons on the legs, proboscis, wings and ovipositor of Drosophila express gustatory receptors(1,2), ion channels(3-6), and ionotropic receptors(7) that are involved in the detection of volatile and non-volatile sensory cues. These neurons directly contact tastants such as food, noxious substances and pheromones and therefore influence many complex behaviors such as feeding, egg-laying and mating. Electrode recordings and calcium imaging have been widely used in insects to quantify the neuronal responses evoked by these tastants. However, electrophysiology requires specialized equipment and obtaining measurements from a single taste sensillum can be technically challenging depending on the cell-type, size, and position. In addition, single neuron resolution in Drosophila can be difficult to achieve since taste sensilla house more than one type of chemosensory neuron. The live calcium imaging method described here allows responses of single gustatory neurons in live flies to be measured. This method is especially suitable for imaging neuronal responses to lipid pheromones and other ligand types that have low solubility in water-based solvents.
