Developing a fast electrochemical aptasensor method for the quantitative detection of penicillin G residue in milk with high sensitivity and good anti-fouling ability

Abstract:<p>A folding-type electrochemical aptasensor was developed for the quantitative detection of penicillin G (Pen-G) residue in milk. The sensor signaling is based on the target binding-induced a change in the probe conformation. The change can drive the MB labels close to the electrode surface, thus producing a big increase in the detection signal. The effect of matrix on sensor response was systematically investigated. The results show that the sensor can respectively produce 23.4 ± 2.8% and 95.2 ± 3.2% of detection signal enhancement (SE) change in the presence of Pen-G concentrations of 1.0 nM and 0.5 mM in buffer. The corresponding limit of detection (LOD) is ~0.3 nM and the response equilibrium time is 90 s. When validated for the quantitative detection of practical milk samples, the sensor performed well with LOD of ~1.7 nM and whole measurement time of &lt;30 min. In addition, both the accuracy and the precision were good with the mean recoveries of 80.2–92.3% and the relative standard deviation (RSD) of &lt;10.0%. The proposed sensing method highlighted high sensitivity and good anti-fouling ability with also good selectivity, quick response, and no sample pretreatment, which can be directly used for the rapid quantitative detection of Pen-G residues in practical milk samples.</p>
